金雀異黃素對(duì)PCOS大鼠卵巢和子宮組織中P450arom mRNA的作用
[Abstract]:Objective: To observe the effect of genistein (GEN) on the key enzyme of estrogen and androgen conversion (P450arom) in ovarian tissues of rats by establishing hyperandrogenism animal model of polycystic ovary syndrome (PCOS). Methods: According to insulin (INS) combined with HCG method, the experimental animal model was established and divided into negative control group, model group with low GEN and middle GEN. The estrogen group was given diethylstilbestrol 0.5 mg/kg for 15 days, and the estrous cycle was observed for 10 days. After the experiment, the blood, uterus and ovary of rats were taken out. The levels of estradiol (E2) and testosterone (T) in serum were measured by ELISA, and the E2/T ratio was calculated. Results: Compared with the model group, the levels of serum E2 in the middle and high dose group and estrogen group increased significantly (P 0.05); the levels of serum testosterone in each GEN dose group decreased, the difference was not significant (P 0.05); the E2/T ratio in each GEN dose group and estrogen group increased significantly (P 0.05). (P 0.05). Compared with the model group, the level of P450 AROM m RNA in ovary and uterus of rats in high dose group decreased significantly (P 0.05), the difference was significant; the level of P450 AROM m RNA in ovary and uterus of rats in middle and low dose group also showed a downward trend, the difference was not significant (P 0.05). Conclusion: Different doses of GEN can increase the ratio of E2/T in serum of rats to supplement aromatase. High dose of genistein (20 mg / (kg body weight)) could significantly increase the expression of P450arom m RNA in ovaries and uterus of PCOS rats, and the effect was similar to that of estrogen.
