12種microRNA在卵巢癌中的表達及臨床意義

【摘要】:目的利用公共數(shù)據(jù)庫分析卵巢癌組織中12種micro RNA(mi R)的表達及其臨床意義。方法篩選在GSE14407數(shù)據(jù)集和TCGA數(shù)據(jù)庫中均具有表達信息的micro RNA,得到mi R-10B、mi R-1244、mi R-622、mi R-21、mi R-503、Let-7D、mi R-155、mi R-30C、mi R-17、mi R-101-1、mi R-186以及mi R-770共12種micro RNA,在這12種micro RNA中尋找在卵巢癌(卵巢癌組)和癌旁組織(癌旁組)中表達有差異的基因。利用在TCGA數(shù)據(jù)庫中篩選得到的505例卵巢癌患者數(shù)據(jù),年齡按照中位數(shù)分為高年齡(≥59歲)組和低年齡(59歲)組;臨床分期分為早期組(Ⅰ期+Ⅱ期)、晚期組(Ⅲ期+Ⅳ期)及未知;腫瘤分級分為低級別(G1+G2)組和高級別(G3+G4)組;病發(fā)位置分為單側(cè)組、雙側(cè)組;腫瘤殘余分為10 mm組、≥10 mm組;micro RNA按照表達量的中位數(shù)分為高表達組和低表達組,進行Kaplan-Meier單因素生存分析和多因素Cox回歸分析。結(jié)果癌旁組Let-7D和mi R-101-1表達水平高于卵巢癌組,而mi R-155和mi R-770表達水平低于卵巢癌組(均P0.05)。生存分析顯示,年齡≥59歲、臨床分期(Ⅲ+Ⅳ)期和Let-7D低表達的患者生存情況較差(P0.05)。Let-7D低表達是卵巢癌患者生存的獨立危險因素。結(jié)論 Let-7D的表達與卵巢癌預后相關,可能是卵巢癌預后的獨立影響指標。
