PDCD5在子宮內(nèi)膜癌中的表達及臨床意義

【摘要】:目的:本文旨在通過檢測PDCD5 (programmed cell death 5,程序性細胞死亡因子5)在子宮內(nèi)膜癌(Endometrial carcinoma, EC)組織中的表達水平,結(jié)合臨床資料,并分析其意義,初步探討PDCD5的表達與子宮內(nèi)膜癌發(fā)生的相關(guān)性。方法:收集2012年5月至2013年2月于濱州醫(yī)學院附屬醫(yī)院婦科行婦科手術(shù),且均經(jīng)病理學檢查證實為EC為研究對象,總標本例數(shù)n=60;另隨機選取同期行婦科手術(shù)中所取得子宮內(nèi)膜不典型增生(endometrial atypical hyperplasia, EAH)組織(n=28)和正常增生期子宮內(nèi)膜(normal proliferative endometrium, NPE)組織(n=28)作為對照。分別采用實時熒光定量PCR(Realtime fluores-cence quantitative PCR, Real time PCR)和免疫組織化學法(Immunohistochemical, IHC)檢測PDCD5 mRNA和蛋白的表達水平,并分析其與子宮內(nèi)膜癌病理特征的關(guān)系。本實驗所遵守的標準為濱州醫(yī)學院附屬醫(yī)院倫理委員會所制定的相關(guān)倫理學標準。結(jié)果:子宮內(nèi)膜癌中PDCD5 mRNA的表達明顯低于對照組,且差異有統(tǒng)計學意義,P≤0.05。PDCD5蛋白在EC組織中的陽性表達率為45.0%,顯著低于EAH及NPE組織中的陽性表達率(P≤0.05)。而且結(jié)果顯示PDCD5的表達水平和EC患者的臨床分期、組織學分級、肌層浸潤以及有無淋巴結(jié)轉(zhuǎn)移有相關(guān)性(均P≤0.05),而與子宮內(nèi)膜癌患者的年齡未發(fā)現(xiàn)明顯的關(guān)聯(lián)性(P0.05)。結(jié)論:采用實時熒光定量PCR和免疫組織化學技術(shù)檢測子宮內(nèi)膜癌組織中PDCD5的表達,發(fā)現(xiàn)PDCD5在子宮內(nèi)膜癌組織中的表達較對照組織中的表達降低甚至缺失。且PDCD5的表達水平與子宮內(nèi)膜癌的臨床病理參數(shù)密切相關(guān):臨床分期越高,PDCD5表達越低;組織學分級越高,PDCD5蛋白的表達越低;肌層浸潤越深,PDCD5表達越低;淋巴結(jié)有轉(zhuǎn)移,PDCD5表達越低;PDCD5表達與年齡無關(guān)。PDCD5有可能成為一種潛在的子宮內(nèi)膜癌的早期診斷分子標記物。
[Abstract]:Objective: to investigate the expression of PDCD5 (programmed cell death 5 and programmed cell death factor 5 (PDCD5 (programmed cell death 5) in endometrial carcinoma (Endometrial carcinoma, EC) and to analyze their clinical significance. To explore the correlation between the expression of PDCD5 and the occurrence of endometrial carcinoma. Methods: from May 2012 to February 2013, gynecological operations were performed in affiliated Hospital of Binzhou Medical College, and were confirmed as EC by pathological examination. Another randomly selected endometrial atypical hyperplasia (endometrial atypical hyperplasia, EAH) tissue (nm28) and normal proliferative endometrial (normal proliferative endometrium, NPE) tissue (nm28) obtained during gynecological surgery were used as control group. The expression of PDCD5 mRNA and protein was detected by real-time fluorescence quantitative PCR (Realtime fluores-cence quantitative PCR, Real time PCR) and immunohistochemical (Immunohistochemical, IHC), and the relationship between the expression of PDCD5 mRNA and the pathological features of endometrial carcinoma was analyzed.
