【摘要】：目的觀察宮頸鱗癌組織中β-連環(huán)蛋白(β-catenin)、E-鈣黏蛋白(E-cadherin)、磷酸酶及張力蛋白同源(PTEN)蛋白的表達(dá)變化,并探討其臨床意義。方法 82例份宮頸鱗癌組織標(biāo)本為觀察組,32例份宮頸上皮內(nèi)瘤變(CIN)組織標(biāo)本為CIN組,30例份正常宮頸組織標(biāo)本為正常組。采用免疫組化SP法進(jìn)行檢測(cè)各組β-catenin、Ecadherin及PTEN蛋白,表達(dá)與宮頸癌臨床病理參數(shù)的關(guān)系,分析β-catenin、E-cadherin及PTEN蛋白的相關(guān)性,采用ROC曲線判斷β-catenin、E-cadherin及PTEN蛋白在宮頸癌診斷中的靈敏度及特異度。結(jié)果觀察組β-catenin陽(yáng)性表達(dá)率高于CIN組和對(duì)照組,E-cadherin、PTEN蛋白陽(yáng)性表達(dá)率均低于CIN組和對(duì)照組(P均0.05)。CIN組β-catenin陽(yáng)性表達(dá)率高于對(duì)照組,E-cadherin、PTEN蛋白陽(yáng)性表達(dá)率均低于對(duì)照組(P均0.05)。β-catenin表達(dá)與宮頸鱗癌組織學(xué)分級(jí)、臨床分期及淋巴結(jié)轉(zhuǎn)移相關(guān)(P均0.05);E-cadherin、PTEN蛋白表達(dá)與宮頸鱗癌組織學(xué)分級(jí)、臨床分期、浸潤(rùn)深度與淋巴結(jié)轉(zhuǎn)移相關(guān)(P均0.05)。β-catenin診斷宮頸鱗癌的靈敏度、特異度分別為89.02%、87.10%,E-cadherin診斷宮頸鱗癌的靈敏度、特異度分別為85.37%、83.87%,PTEN蛋白診斷宮頸鱗癌的靈敏度、特異度分別為84.15%、85.48%。宮頸鱗癌組織中β-catenin表達(dá)與E-cadherin、PTEN呈負(fù)相關(guān)(r分別為-0.736,-0.702;P0.05),E-cadherin表達(dá)與PTEN蛋白表達(dá)呈正相關(guān)(r=0.694,P0.05)。結(jié)論宮頸鱗癌組織中β-catenin高表達(dá),E-cadherin、PTEN蛋白低表達(dá)。β-catenin、E-cadherin及PTEN蛋白診斷宮頸鱗癌的敏感度和特異度均較高。β-catenin、E-cadherin及PTEN在宮頸鱗癌的發(fā)生發(fā)展中可能存在相互作用。
[Abstract]:Objective to observe the expression of 尾 -catenin (尾 -catenin) E-cadherin (E-cadherin), phosphatase and tensin homologous (PTEN) protein in cervical squamous cell carcinoma (SCC) and to explore its clinical significance. Methods Eighty-two specimens of cervical squamous cell carcinoma were collected from 32 patients with cervical intraepithelial neoplasia (CIN) and 30 normal cervix tissues from CIN group. Immunohistochemical SP method was used to detect the expression of 尾 -cateninine E-cadherin and PTEN protein, and the relationship between the expression of 尾 -cateninine E-cadherin and PTEN protein was analyzed. The sensitivity and specificity of 尾 -cateninine E-cadherin and PTEN protein in the diagnosis of cervical cancer were evaluated by ROC curve. Results the positive expression rate of 尾 -catenin in observation group was higher than that in CIN group and control group. The positive expression rate of 尾 -catenin protein in observation group was lower than that in CIN group and control group (P 0.05). The positive expression rate of 尾 -catenin protein in cin group was higher than that in control group. And histological grade of cervical squamous cell carcinoma, The expression of E-cadherin PTEN protein was correlated with the histological grade, clinical stage, depth of invasion and lymph node metastasis (P 0.05). The sensitivity of 尾 -catenin in the diagnosis of cervical squamous cell carcinoma was significantly correlated with the clinical stage and lymph node metastasis (P 0.05). The sensitivity of E-cadherin in the diagnosis of cervical squamous cell carcinoma was 89.02 and 87.10, and the specificity was 85.37 and 83.87, respectively. The specificity of PTEN protein in the diagnosis of cervical squamous cell carcinoma was 84.15 and 85.48, respectively. There was a negative correlation between the expression of 尾 -catenin and E-cadherin in cervical squamous cell carcinoma (r = -0.736- 0.702P respectively). The expression of E-cadherin was positively correlated with the expression of PTEN protein (r = 0.694P 0.05). Conclusion the high expression of 尾 -catenin and the low expression of E-cadherin PTEN protein in cervical squamous cell carcinoma. The sensitivity and specificity of 尾 -cateninine E-cadherin and PTEN protein in the diagnosis of cervical squamous cell carcinoma are higher. There may be interaction between 尾 -catenin E-cadherin and PTEN in the occurrence and development of cervical squamous cell carcinoma.
