【摘要】：目的 :研究GATAD1在子癇前期患者胎盤組織中的表達(dá)及其意義。方法 :29例由美國(guó)Mayo Clinic婦產(chǎn)科臨床醫(yī)師收集的胎盤組織,其中早孕正常胎盤8例(分娩孕周為7~12周,平均為10.2周),晚孕正常胎盤14例(分娩孕周為35~41周,平均為39.5周),晚孕子癇前期胎盤7例(分娩孕周為31~41周,平均為34.6周)。采用RT-PCR技術(shù)、蛋白印跡(Western blot)法和免疫組化檢測(cè)3組孕婦胎盤組織中GATAD1的m RNA與蛋白表達(dá)水平差異。結(jié)果:晚孕正常胎盤組織中GATAD1的m RNA與蛋白表達(dá)水平均顯著高于早孕正常胎盤組織。子癇前期組患者胎盤組織中GATAD1的m RNA與蛋白表達(dá)水平均顯著低于相應(yīng)孕周正常妊娠組。結(jié)論:GATAD1表達(dá)的下降與子癇前期的發(fā)病相關(guān)。
[Abstract]:Objective: to study the expression and significance of GATAD1 in placenta of preeclampsia. Methods Twenty nine cases of placental tissue were collected by clinical doctors of gynecology and obstetrics at the Mayo Clinic in the United States. Among them, 8 cases were normal placenta of early pregnancy (7 weeks of gestation, averaging 10.2 weeks), 14 cases of normal placenta of late pregnancy (35 weeks and 41 weeks of gestation). The average gestational age was 39.5 weeks and the placenta of preeclampsia was found in 7 cases (the gestational age was 31 weeks, with an average of 34.6 weeks). The mRNA and protein expression levels of GATAD1 in placenta of pregnant women were detected by RT-PCR, Western blot and immunohistochemistry. Results: the mRNA and protein expression levels of GATAD1 in normal placenta of late pregnancy were significantly higher than those in normal placenta of early pregnancy. The expression of GATAD1 mRNA and protein in placenta of preeclampsia group was significantly lower than that of normal gestational group. Conclusion the decrease of the expression of Gata D1 is associated with preeclampsia.
