E7蛋白在HPV16變異體持續(xù)感染患者宮頸組織中的表達(dá)

【摘要】:目的:探索HPV16亞洲變異體E6 T178G及歐洲變異體E6 T350G,A442C與其他變異體致宮頸病變及機(jī)制的異同。方法:選取浦東新區(qū)人民醫(yī)院門診或住院患者中HPV16型單一陽(yáng)性、宮頸細(xì)胞學(xué)正常的300例研究對(duì)象,予宮頸液基薄層細(xì)胞學(xué)檢查(Thinprep cytologic test,TCT)采集宮頸脫落細(xì)胞,對(duì)其E2、E6、E7基因序列進(jìn)行檢測(cè);1年后復(fù)查TCT、HPV,持續(xù)感染者再次基因測(cè)序明確HPV16變異體分型,并對(duì)持續(xù)感染者進(jìn)行陰道鏡活檢后病理檢查,對(duì)所有患者均用SP免疫組化法檢測(cè)E7蛋白表達(dá)水平。結(jié)果:HPV16變異體持續(xù)感染1年后,292例成功測(cè)序的患者中,259例慢性宮頸炎,32例宮頸上皮內(nèi)瘤變Ⅰ級(jí)(CINI),1例宮頸上皮內(nèi)瘤變Ⅰ級(jí)(CINⅡ)。E7蛋白在各種變異體感染的患者宮頸組織中均有表達(dá),其在亞洲變異體及歐洲變異體感染組與其他變異體感染組之間表達(dá)無(wú)明顯差異(P0.05)。結(jié)論:E7蛋白可能在HPV16變異體致宮頸病變的早期階段即發(fā)揮作用,但E7蛋白可能并不是HPV16不同變異體致宮頸病變機(jī)制不同的參考指標(biāo)。
[Abstract]:Objective: to explore the similarities and differences of cervical lesions caused by HPV16 Asian variant E6T178G and European variant E6T350GA442C with other variants. Methods: cervical exfoliated cells were collected from 300 patients with single HPV16 positive and normal cervical cytology in the outpatient or inpatient of Pudong New area people's Hospital. The cervical exfoliated cells were collected by Thinprep cytologic test TCT (Thinprep cytologic test TCT). The sequence of E2OE6 E7 gene was detected, and a year later, the HPV16 variant was determined by resequencing of the HPV16 gene, and the colposcopy biopsy was performed on the HPV16 mutation in the patients with persistent infection after colposcopy biopsy, and the HPV16 mutation was determined by re-sequencing of the HPV16 gene. The expression of E7 protein was detected by SP immunohistochemical method in all patients.
