雨生紅球藻在子癇前期疾病中的應用

【摘要】:目的探討雨生紅球藻對L-亞硝酸左旋精氨酸甲酯(L-NAME)誘導的子癇前期模型大鼠病理組織改變和氧化應激的保護作用及機制。方法采用L-亞硝酸左旋精氨酸甲酯(L-NAME)誘導產生子癇前期孕鼠,隨機分為空白組(n=20)、L-NAME建模組(n=20)和雨生紅球藻組(n=20)。受孕D5,雨生紅球藻組大鼠灌胃1.6g/kg bw.d雨生紅球藻;D10建模組和雨生紅球藻組每天皮下注射L-NAME 125 mg/kg bw·d。檢測血液中MDA、SOD、NOS;HE和Masson染色分析胎盤組織病理改變;免疫組化分析NF-κB(nuclear factor kappa B)、ROCK II(Rho-associated protein kinase II)、HO-1(heme oxygenase)、caspase 3蛋白表達變化。結果 L-NAME誘導大鼠產生典型的子癇前期表現,如血壓、尿蛋白增高;血液中的MDA、內皮素(endothelin,ET)增高。而經雨生紅球藻處理后,大鼠血壓顯著降低,尿蛋白減少,血液中的MDA和內皮素下降,同時雨生紅球藻明顯下調了胎盤組織中的NF-κB、ROCK Ⅱ、caspase 3,增加了HO-1的表達。胎盤組織中的滋養(yǎng)細胞數減少,纖維蛋白沉積減少,基底膜增厚現象改善。結論雨生紅球藻能有效減輕子癇前期大鼠的氧化應激損傷及病理組織改變,對子癇前期疾病有一定的預防及治療作用。
[Abstract]:Objective to investigate the protective effect and mechanism of Haematococcus pluvialis on pathological changes and oxidative stress in rats with preeclampsia induced by L-arginine methyl nitrite (L-NAME). Methods pregnant rats with preeclampsia induced by L-arginine methyl nitrite (L-NAME) were randomly divided into two groups: control group (nr-20), L-NAME model group (nong20) and erythrocystis pluvialis group (nd20). Pregnant D5, 1.6g/kg bw.d mg/kg D10 and L-NAME 125 mg/kg BW were injected subcutaneously into the model group and the control group respectively. The histopathological changes of placenta were detected by HE and Masson staining, and the expression of HO-1 (heme oxygenase) caspase 3 was analyzed by immunohistochemistry. Results L-NAME induced typical preeclampsia in rats, such as increased blood pressure and urine protein, increased MDAand endothelin et in blood. However, the blood pressure, urine protein, MDA and endothelin in the blood of rats were significantly decreased after treated with Haematococcus pluvialis, while the expression of HO-1 in placenta was significantly down-regulated by Haematococcus pluvialis, and NF- 魏 Brock 鈪,
