【摘要】：目的探討子癇前期胎盤組織中miR-26b差異表達及其臨床意義。方法收集子癇患者與健康人胎盤組織各30例,RT-qPCR和Western blotting檢測組織中miR-26b與Smad1的表達;轉(zhuǎn)染miR-26b的mimics到HTR-8/SVneo細胞中,MTT法比較轉(zhuǎn)染組、NC對照組細胞增殖情況,流式細胞術(shù)檢測各組細胞周期情況;同時RT-qPCR、Western blotting檢測各組miR-26b和Smad1的表達情況,并運用雙熒光素酶報告檢測試驗驗證miR-26b與靶標Smad1之間靶向關(guān)系。結(jié)果 miR-26b在子癇前期胎盤組織中高表達,且Smad1的表達與miR-26b的表達相反;滋養(yǎng)層細胞中miR-26b過表達會抑制細胞增殖(P0.01),并將細胞阻滯在G0/G1期。miR-26b過表達會降低Smad1 mRNA與蛋白的表達(P0.01)。雙熒光素酶報告基因?qū)嶒炞C明Smad1是miR-26b的直接靶點。結(jié)論 miR-26b可能通過靶向Smad1調(diào)控HTR-8/SVneo細胞的增殖與周期。
[Abstract]:Objective to investigate the differential expression of miR-26b in placenta of preeclampsia and its clinical significance. Methods the expression of miR-26b and Smad1 in 30 cases of placental tissues from patients with eclampsia and 30 cases of healthy subjects were detected by RT-PCR and Western blotting, and the proliferation of NC cells in the transfected group was compared by mimics transfected with miR-26b into HTR-8 / SVneo cells. The expression of miR-26b and Smad1 in each group was detected by RT-qPCRG Western blotting, and the targeting relationship between miR-26b and target Smad1 was verified by double-luciferase report assay. Results the expression of miR-26b in placenta of preeclampsia was higher than that of miR-26b, and the overexpression of miR-26b in trophoblast cells inhibited the proliferation of P0.01and blocked the cells in G0 / G1 phase. MiR-26b overexpression decreased the expression of Smad1 mRNA and protein. Double luciferase reporter gene experiment showed that Smad1 was a direct target of miR-26b. Conclusion miR-26b may regulate the proliferation and cell cycle of HTR-8 / SVneo cells by targeting Smad1.
