【摘要】：目的探討子癇前期胎盤(pán)組織中miR-26b差異表達(dá)及其臨床意義。方法收集子癇患者與健康人胎盤(pán)組織各30例,RT-qPCR和Western blotting檢測(cè)組織中miR-26b與Smad1的表達(dá);轉(zhuǎn)染miR-26b的mimics到HTR-8/SVneo細(xì)胞中,MTT法比較轉(zhuǎn)染組、NC對(duì)照組細(xì)胞增殖情況,流式細(xì)胞術(shù)檢測(cè)各組細(xì)胞周期情況;同時(shí)RT-qPCR、Western blotting檢測(cè)各組miR-26b和Smad1的表達(dá)情況,并運(yùn)用雙熒光素酶報(bào)告檢測(cè)試驗(yàn)驗(yàn)證miR-26b與靶標(biāo)Smad1之間靶向關(guān)系。結(jié)果 miR-26b在子癇前期胎盤(pán)組織中高表達(dá),且Smad1的表達(dá)與miR-26b的表達(dá)相反;滋養(yǎng)層細(xì)胞中miR-26b過(guò)表達(dá)會(huì)抑制細(xì)胞增殖(P0.01),并將細(xì)胞阻滯在G0/G1期。miR-26b過(guò)表達(dá)會(huì)降低Smad1 mRNA與蛋白的表達(dá)(P0.01)。雙熒光素酶報(bào)告基因?qū)嶒?yàn)證明Smad1是miR-26b的直接靶點(diǎn)。結(jié)論 miR-26b可能通過(guò)靶向Smad1調(diào)控HTR-8/SVneo細(xì)胞的增殖與周期。
[Abstract]:Objective to investigate the differential expression of miR-26b in placenta of preeclampsia and its clinical significance. Methods the expression of miR-26b and Smad1 in 30 cases of placental tissues from patients with eclampsia and 30 cases of healthy subjects were detected by RT-PCR and Western blotting, and the proliferation of NC cells in the transfected group was compared by mimics transfected with miR-26b into HTR-8 / SVneo cells. The expression of miR-26b and Smad1 in each group was detected by RT-qPCRG Western blotting, and the targeting relationship between miR-26b and target Smad1 was verified by double-luciferase report assay. Results the expression of miR-26b in placenta of preeclampsia was higher than that of miR-26b, and the overexpression of miR-26b in trophoblast cells inhibited the proliferation of P0.01and blocked the cells in G0 / G1 phase. MiR-26b overexpression decreased the expression of Smad1 mRNA and protein. Double luciferase reporter gene experiment showed that Smad1 was a direct target of miR-26b. Conclusion miR-26b may regulate the proliferation and cell cycle of HTR-8 / SVneo cells by targeting Smad1.
