RRM1和RRM2表達(dá)水平與宮頸癌化療的相關(guān)性研究

【摘要】:目的核糖核苷酸還原酶(Ribonucleotide reductase,RR)是細(xì)胞內(nèi)DNA復(fù)制和損傷修復(fù)中的限速酶,在細(xì)胞增殖以及腫瘤增殖和浸潤(rùn)轉(zhuǎn)移中起著重要作用。宮頸癌在女性高發(fā)惡性腫瘤中位于第二位,化療是宮頸癌治療的一種重要手段。RR是一些抗腫瘤藥物的作用靶點(diǎn),吉西他濱(Gemcitabine,GEM)是目前最有效且應(yīng)用最廣泛的RR抑制劑。本研究通過檢測(cè)宮頸癌組織和細(xì)胞中RR酶活性及亞基表達(dá)水平,探討RR在宮頸癌發(fā)生、發(fā)展的可能作用;進(jìn)一步研究吉西他濱與卡鉑(Carboplatin, CBDCA)聯(lián)合用藥對(duì)宮頸癌細(xì)胞的作用效果。 方法采用實(shí)時(shí)熒光定量PCR(qRT-PCR)、Western Blotting和CDP還原法分別檢測(cè)宮頸癌組織及癌旁正常組織中RR亞基表達(dá)及酶活性,CCK-8法檢測(cè)宮頸癌細(xì)胞系SiHa、CaSki對(duì)GEM、CBDCA單藥及合藥的敏感性,EdU法檢測(cè)GEM對(duì)宮頸癌細(xì)胞DNA合成的影響,Western Blotting和免疫熒光兩種方法檢測(cè)藥物對(duì)宮頸癌細(xì)胞的DNA損傷作用,流式細(xì)胞儀檢測(cè)GEM、CBDCA單藥及合藥時(shí)細(xì)胞凋亡情況,并基于Chou-Talalay法計(jì)算合用指數(shù)(CI)以分析GEM與CBDCA合用對(duì)宮頸癌細(xì)胞是否具有協(xié)同作用。 結(jié)果宮頸癌組織中的RRM1和RRM2mRNA水平較癌旁正常組織中的表達(dá)水平明顯升高(P0.0001),類似地,RR酶活和亞基蛋白表達(dá)水平在宮頸癌組織中的水平也上調(diào)。GEM能抑制宮頸癌細(xì)胞DNA合成,CBDCA能誘導(dǎo)DNA損傷,,GEM與CBDCA聯(lián)合作用對(duì)宮頸癌細(xì)胞具有協(xié)同毒性作用(log10[CI]0),GEM與CBDCA合用能促進(jìn)誘導(dǎo)宮頸癌細(xì)胞DNA損傷產(chǎn)生和促進(jìn)細(xì)胞凋亡率增加。 結(jié)論核糖核苷酸還原酶在宮頸癌中的表達(dá)上調(diào),細(xì)胞實(shí)驗(yàn)表明RR抑制劑GEM與宮頸癌一線化療藥物CBDCA合用對(duì)宮頸癌細(xì)胞產(chǎn)生協(xié)同毒性作用,協(xié)同作用是通過抑制DNA合成和促進(jìn)細(xì)胞凋亡產(chǎn)生的。本研究可能為GEM聯(lián)合CBDCA在宮頸癌的臨床治療應(yīng)用提供實(shí)驗(yàn)依據(jù)。
[Abstract]:Objective ribonucleotide reductase ( RR ) is a limiting enzyme in DNA replication and repair of cervical cancer , plays an important role in cell proliferation and tumor proliferation and invasion .
To further study the effect of gemcitabine and carboplatin ( CBDCA ) on cervical cancer cells .
Methods The expression of RR subunits in cervical cancer tissues and adjacent normal tissues was detected by real - time fluorescence quantitative PCR ( qRT - PCR ) , Western Blotting and CDP reduction . The effects of GEM and CBDCA alone on DNA synthesis of cervical cancer cells were detected by the method of Western Blotting and CDP . The effect of GEM on DNA synthesis of cervical cancer cells was detected by the method of Western Blotting and Immunofluorescence .
Results The levels of RRM1 and RRM2mRNA in cervical cancer tissues were significantly higher than those in normal tissues ( P0.05 ) .
Conclusion The expression of ribonucleotide reductase in cervical cancer is up - regulated . The results suggest that the combination of the RR inhibitor GEM and the first - line chemotherapy drug CBDCA has a synergistic effect on cervical cancer cells . The synergistic effect is the inhibition of DNA synthesis and the promotion of apoptosis . This study may provide experimental basis for the application of GEM and CBDCA in the clinical treatment of cervical cancer .
