缺氧通過刺激白介素8分泌促進宮頸癌細(xì)胞增殖

【摘要】:目的:探討缺氧與白介素8對宮頸癌細(xì)胞的生存活力及凋亡是否具有調(diào)節(jié)作用及其可能的機制。 方法:免疫組化分析宮頸癌和宮頸炎組織中缺氧誘導(dǎo)因子(hypoxia inducible factor-1α,HIF-1α)、白介素8(interleukin8,IL-8)及其受體CXCR1和CXCR2的表達差異,采用磺酰羅丹明B(Sulforhodamine B,SRB)比色法及流式細(xì)胞術(shù)分析缺氧和白介素8對宮頸癌HeLa與SiHa細(xì)胞生存活力及凋亡的影響。結(jié)果:宮頸癌組織中HIF-1α、IL-8和CXCR1的表達水平明顯高于宮頸炎 組織,缺氧可以刺激HeLa與SiHa細(xì)胞IL-8的分泌并增強其受體CXCR1和CXCR2的表達,重組人IL-8細(xì)胞因子可以促進HeLa與SiHa細(xì)胞的生存活力并降低凋亡率。在1%氧氣條件下培養(yǎng),HeLa與SiHa細(xì)胞的生存活力和凋亡水平增加,并且抗人IL-8中和性抗體可以部分阻斷缺氧對HeLa和SiHa細(xì)胞活力的促進作用。 結(jié)論:缺氧可以誘導(dǎo)宮頸癌細(xì)胞分泌IL-8,,IL-8可以自分泌的方式增強宮頸癌細(xì)胞的生存活力,進而參與宮頸癌的發(fā)病。
[Abstract]:Objective: to investigate whether hypoxia and interleukin-8 can regulate the viability and apoptosis of cervical cancer cells and their possible mechanisms. Methods: the hypoxia-inducible factor-1 偽 -HIF-1 偽 in cervical carcinoma and cervicitis was analyzed by immunohistochemistry. The expression of IL-8 and its receptors CXCR1 and CXCR2 were different. The effects of hypoxia and interleukin-8 (IL-8) on the viability and apoptosis of cervical cancer HeLa and SIHA cells were analyzed by using sulfonyl rhodamine BSulforhodamine BHS-SRB colorimetry and flow cytometry. Results: the expression levels of HIF-1 偽, IL-8 and CXCR1 in cervical carcinoma were significantly higher than those in cervicitis. Hypoxia stimulated the secretion of IL-8 and enhanced the expression of CXCR1 and CXCR2 in HeLa and SIHA cells. Recombinant human IL-8 cytokines can promote the viability of HeLa and SIHA cells and decrease the apoptosis rate. The survival activity and apoptosis level of HeLa and SIHA cells cultured in 1% oxygen were increased. And anti-human IL-8 neutralizing antibody can partially block the effect of hypoxia on the activity of HeLa and SIHA cells. Conclusion: hypoxia can induce cervical cancer cells to secrete IL-8IL-8 to enhance the viability of cervical cancer cells in an autocrine manner. Then participate in the pathogenesis of cervical cancer.
