【摘要】：目的探討子宮內(nèi)膜異位癥(endometriosis,EMs)不孕患者腹腔液中的微生物菌群及其腫瘤壞死因子α(TNF-α)和白介素37(IL-37)水平的變化。方法采集內(nèi)膜異位癥不孕組(T)29例和對照組(C)19例女性的腹腔液(peritoneal fluid,PF)標(biāo)本,分別進(jìn)行基因組DNA提取后Ion Torrent PGM平臺測序檢測微生物菌群和酶聯(lián)免疫吸附試驗(yàn)(ELISA)檢測IL-37及TNF-α水平。結(jié)果 EMs不孕患者和對照組女性PF中均發(fā)現(xiàn)了多種菌群存在,在菌門水平上主要分為變形菌門Proteobacteria、厚壁菌門Firmicutes、擬桿菌門Bacteriodetes、放線菌門Actinobacteria、梭桿菌門Fusobacteria,其中Proteobacteria為主要菌群,占60%以上的序列,軟壁菌門Tenericutes只在對照組中檢出1例,各菌門在兩組間差異無統(tǒng)計(jì)學(xué)意義(P0.05)。EMs不孕婦女腹腔液中TNF-a和IL-37水平分別為(75.87±8.47)pg/ml、(372.03±68.02)pg/ml,高于對照組(57.76±8.29)pg/ml和(261.37±52.78)pg/ml(P0.05);EMs不孕組Ⅲ~Ⅳ期TNF-a的水平為(83.47±4.73)pg/ml,顯著高于Ⅰ~Ⅱ期(74.29±8.26)pg/ml(P0.05),IL-37的水平在二者中差異無統(tǒng)計(jì)學(xué)意義(P0.05);內(nèi)膜異位癥不孕組中兩者存在正相關(guān)關(guān)系(r=0.569,P0.001),對照組中兩者不存在相關(guān)關(guān)系(r=-0.247,P=0.522)。結(jié)論 EMs不孕患者和對照組女性腹腔液中存在多種菌群,EMs不孕組中TNF-a和IL-37含量增高可能在EMs不孕癥的發(fā)生機(jī)制中起到重要作用。
[Abstract]:Objective to investigate the changes of microflora, tumor necrosis factor 偽 (TNF- 偽) and interleukin-37 (IL-37) in peritoneal fluid of infertile patients with endometriosis. Methods the peritoneal fluid of 29 infertile women with endometriosis and 19 female controls with endometriosis were collected, and the levels of IL-37 and TNF- 偽 were detected by Ion Torrent PGM platform sequencing after genomic DNA extraction. The levels of IL-37 and TNF- 偽 were detected by enzyme linked immunosorbent assay (Elisa). Results A variety of microflora was found in female PF of infertile patients and control group, which was divided into Proteobacteria, Firmicutes, Bacteriodetes, Actinobacteria, Fusobacteria, Proteobacteria, among which Proteobacteria was the main group. Tenericutes was detected only in 1 case in the control group. There was no significant difference between the two groups in the levels of TNF-a and IL-37 in the peritoneal fluid of infertile women (75.87 鹵8.47 pg / ml, 372.03 鹵68.02), which were higher than those in the control group (57.76 鹵8.29)pg/ml) and the control group (261.37 鹵52.78).
