SDF-1和CXCR4在宮頸癌組織中的表達及臨床意義

【摘要】:目的:大量研究發(fā)現(xiàn)基質(zhì)細胞衍生因子-1(SDF-1)和趨化因子受體4(CXCR4)在多種腫瘤的發(fā)生發(fā)展、浸潤轉(zhuǎn)移中發(fā)揮著重要作用,,本文旨在探討SDF-1和CXCR4兩種蛋白在宮頸癌中的表達及其臨床意義。 方法:應用免疫組織化學染色法,檢測80例子宮頸癌,20例正常子宮頸組織中SDF-1和CXCR4蛋白的表達情況;并通過分析比較兩者與宮頸癌患者的年齡、病理分級、臨床分期、淋巴結(jié)轉(zhuǎn)移等方面的關系;研究CXCR4、SDF-1與宮頸癌的發(fā)生、發(fā)展有無關系,并分析兩者之間有無相關性;為宮頸癌的診斷治療提供一種新的分子靶向治療的方法。 結(jié)果:1.SDF-1蛋白在正常宮頸組織及宮頸癌組織中的表達、SDF-1的表達與宮頸癌的臨床病理因素之間的關系 SDF-1蛋白在正常宮頸組織中陽性率為5%(1/20),在宮頸癌組織中表達的陽性率為55%(44/80),二者陽性率的差別有統(tǒng)計學意義(P0.05)。 SDF-1蛋白的表達在宮頸癌臨床分期≥Ⅱa期組的陽性率較臨床分期Ⅱa期組明顯增加(P0.05);有淋巴結(jié)轉(zhuǎn)移組的陽性率較無淋巴結(jié)轉(zhuǎn)移組明顯增加(P0.05);病理分級Ⅰ、Ⅱ、Ⅲ級組的陽性率的差別有統(tǒng)計學意義(P0.05),病理分級Ⅲ級組的陽性率較Ⅰ、Ⅱ級組明顯增加(P0.01、P0.01),但病理分級Ⅰ、Ⅱ級組的陽性率無統(tǒng)計學差異(P0.05)。 SDF-1蛋白表達的陽性率與宮頸癌患者年齡大小(P0.05)、原發(fā)灶大小(P0.05)、組織類型(P0.05)均無關,無統(tǒng)計學差異。 2. CXCR4蛋白在正常宮頸組織及宮頸癌組織中的表達、CXCR4的表達與宮頸癌的臨床病理因素之間的關系 CXCR4蛋白在正常宮頸組織中陽性率為10%(2/20),在宮頸癌組織中表達的陽性率為52.5%(42/80),二者陽性率的差別有統(tǒng)計學意義(P0.05)。 CXCR4蛋白的表達在宮頸癌有淋巴結(jié)轉(zhuǎn)移組的陽性率較無淋巴結(jié)轉(zhuǎn)移組明顯增加(P0.05);臨床分期≥Ⅱa期組的陽性率較臨床分期Ⅱa期組明顯增(P0.05);原發(fā)灶大小≥4cm組較原發(fā)灶大小4cm組明顯增加(P0.05);病理分級Ⅰ、Ⅱ、Ⅲ級組的陽性率差別有統(tǒng)計學意義(P0.05),病理分級Ⅲ級組的陽性率較Ⅰ、Ⅱ級組明顯增加(P0.01,P0.01),但病理分級Ⅰ、Ⅱ級組的陽性率無統(tǒng)計學差異(P0.05)。 CXCR4蛋白表達的陽性率與宮頸癌患者年齡大小(P0.05)、組織類型(P0.05)均無關,無統(tǒng)計學差異。 3.宮頸癌組織中SDF-1、CXCR4蛋白表達的相關性 宮頸癌中SDF-1和CXCR4的表達均增加,二者呈正相關性(R=0.473)。 結(jié)論:趨化因子CXCR4及其配體SDF-1蛋白的高表達在宮頸癌的惡性增殖、浸潤轉(zhuǎn)移有一定意義,它們之間的這種正相關作用可能與宮頸癌的發(fā)生發(fā)展有一定關系。
[Abstract]:Objective: a large number of studies have found that stromal cell derived factor-1 SDF-1) and chemokine receptor 4 (CXCR4) play an important role in the development, invasion and metastasis of many kinds of tumors. The purpose of this study was to investigate the expression and clinical significance of SDF-1 and CXCR4 proteins in cervical cancer. Methods: immunohistochemical staining method was used to detect the expression of SDF-1 and CXCR4 protein in 20 normal cervical tissues from 80 cases of cervical carcinoma, and to compare the expression of SDF-1 and CXCR4 protein with cervical cancer patients' age, pathological grade and clinical stage. To study the relationship between CXCR4 SDF-1 and cervical cancer, and to analyze the relationship between them, and to provide a new molecular targeted therapy for cervical cancer diagnosis and treatment. Results 1. The expression of SDF-1 protein in normal cervix and cervical carcinoma.
