IL-6和SOCS1在上皮性卵巢癌中的表達及臨床意義

【摘要】:目的:通過檢測正常卵巢、良性卵巢腫瘤和上皮性卵巢癌組織中IL-6(interleukin-6)和SOCS1(suppressor of cytokine signaling 1)的表達情況,探討兩者的表達特點及其在卵巢癌發(fā)生發(fā)展中的相互作用、關(guān)系及臨床意義。方法:采用二步免疫組織化學法檢測105例上皮性卵巢癌組織、40例良性卵巢腫瘤組織和30例正常卵巢組織中IL-6和SOCS1的表達水平,利用SPSS軟件分析兩者在不同卵巢組織中的表達情況、兩者與卵巢癌臨床病理參數(shù)間的關(guān)系以及上皮性卵巢癌中兩者的相關(guān)性。結(jié)果:IL-6主要表達于上皮性卵巢癌的胞漿及間質(zhì)中,SOCS1主要表達于上皮性卵巢癌的胞漿及胞核中,兩者在良性卵巢腫瘤和正常卵巢組織中均呈低表達。IL-6在上皮性卵巢癌、良性卵巢腫瘤和正常卵巢組織中的陽性表達率分別為71.43%、22.50%、16.70%,上皮性卵巢癌組織中IL-6的陽性表達率顯著高于良性卵巢腫瘤和正常卵巢組織(P=0.00),良性卵巢腫瘤和正常卵巢組織中IL-6的陽性表達率相比較差距無統(tǒng)計學意義(P0.05);SOCS1在上皮性卵巢癌、良性卵巢腫瘤和正常卵巢組織中的陽性表達率分別為37.14%、15.00%、13.30%,上皮性卵巢癌組織中SOCS1的陽性表達率明顯高于良性卵巢腫瘤和正常卵巢組織,差異有統(tǒng)計學意義(P=0.004),良性卵巢腫瘤和正常卵巢組織中SOCS1的陽性表達率相比較差距無統(tǒng)計學意義(P0.05)。IL-6的表達與卵巢癌的組織學分級、FIGO分期、腹水和淋巴結(jié)轉(zhuǎn)移具有相關(guān)性(均P0.05),與腫瘤病理類型、患者年齡無相關(guān)性(均P0.05)。SOCS1的表達與卵巢癌的組織學分級具有相關(guān)性(P0.05),而與FIGO分期、腹水、病理類型、患者年齡、淋巴結(jié)轉(zhuǎn)移均無相關(guān)性(均P0.05)。IL-6與SOCS1在上皮性卵巢癌中的表達呈正相關(guān)(r=0.297,P0.05)。結(jié)論:(1)IL-6和SOCS1在良性卵巢腫瘤和正常卵巢組織中均呈低表達,在上皮性卵巢癌中兩者的陽性表達率均升高,且兩者的表達呈正相關(guān),說明IL-6和SOCS1與上皮性卵巢癌密切相關(guān),可能在卵巢癌的發(fā)生發(fā)展中起到重要作用。(2)IL-6的表達與卵巢癌患者多種臨床病理特征有關(guān),與卵巢癌的病情嚴重程度密切相關(guān),其可作為預(yù)測卵巢癌預(yù)后的重要因子。(3)SOCS1作為IL-6的負性調(diào)控因子,在上皮性卵巢癌中的陽性表達率低于IL-6,其中的機制尚不明確,這為探索卵巢癌的發(fā)生發(fā)展提供新的線索,可認為有助于尋找新的卵巢癌的治療方法。
[Abstract]:Objective: to investigate the expression of interleukin-6 and SOCS1(suppressor of cytokine signaling _ 1 in normal, benign and epithelial ovarian carcinomas, and to explore their interaction, relationship and clinical significance in the development of ovarian cancer. Methods: the expression of IL-6 and SOCS1 in 105 cases of epithelial ovarian carcinoma (EOC) and 30 cases of normal ovarian tissues were detected by two step immunohistochemical method, including 40 cases of benign ovarian tumors and 30 cases of normal ovarian tissues. SPSS software was used to analyze their expression in different ovarian tissues, the relationship between them and the clinicopathological parameters of ovarian cancer and the correlation between them in epithelial ovarian carcinoma. Results the cell cytoplasm and interstitial cytoplasm of epithelial ovarian carcinoma were mainly expressed in the cytoplasm and nucleus of epithelial ovarian carcinoma, and both of them were low expressed in benign ovarian tumor and normal ovarian tissue. IL-6 was expressed in epithelial ovarian carcinoma. The positive expression rates of IL-6 in benign ovarian tumor and normal ovarian tissue were 71.43 and 22.50 ~ 16.70, respectively. The positive expression rate of IL-6 in epithelial ovarian cancer was significantly higher than that in benign ovarian tumor and normal ovarian tissue. The positive expression rate of IL-6 in benign ovarian tumor and normal ovarian tissue was significantly higher than that in benign ovarian tumor and normal ovarian tissue. There was no significant difference between the positive rate of IL-6 and the expression of SOCS1 in epithelial ovarian carcinoma. The positive expression rates of SOCS1 in benign ovarian tumors and normal ovarian tissues were 37.14 and 15.00, respectively. The positive expression rate of SOCS1 in epithelial ovarian cancer was significantly higher than that in benign ovarian tumors and normal ovarian tissues. The positive expression rate of SOCS1 in benign ovarian tumors and normal ovarian tissues was not significantly higher than that in normal ovarian tissues. There was no significant difference in the expression of SOCS1 between benign ovarian tumors and normal ovarian tissues. There was no significant difference between the expression of P0.05 and IL-6 and the histological grade and Figo stage of ovarian cancer. There was no correlation between ascites and lymph node metastasis (all P0.05), and there was no correlation between the expression of P0.05).SOCS1 and histological grade of ovarian cancer (P 0.05), but with FIGO stage, ascites, pathological type, age of patients. There was no correlation between lymph node metastasis and P0.05).IL-6 and SOCS1 expression in epithelial ovarian carcinoma. Conclusion the expression of IL-6 and SOCS1 in benign ovarian tumors and normal ovarian tissues is low, and the positive expression rate of both in epithelial ovarian cancer is higher, and the positive correlation between them is positive. The results suggest that IL-6 and SOCS1 are closely related to epithelial ovarian cancer. The expression of IL-6 may play an important role in the occurrence and development of ovarian cancer. The expression of IL-6 is related to many clinicopathological features of ovarian cancer, and is closely related to the severity of ovarian cancer. The positive expression rate of SOCS1 in epithelial ovarian cancer is lower than that of IL-6, which provides a new clue for exploring the occurrence and development of ovarian cancer. It can be considered to be helpful in finding a new treatment for ovarian cancer.
