【摘要】：目的:探索NF2基因蛋白Merlin于青海女性非病變宮頸組織及子宮頸鱗狀細胞癌內的顯示狀況;比較青海漢族、藏族、回族宮頸鱗癌Merlin蛋白的顯示是否存在民族差異性,并且對Merlin蛋白在各民族宮頸鱗癌病患的年紀、FIGO期別、淋巴轉移情況及細胞分化水平等臨床病理特性間的顯示情況進行進一步探討。方法:收集2014年07月至2016年07月因子宮頸鱗癌在青海紅十字醫(yī)院行宮頸癌根治術治療的108例子宮頸鱗癌組織標本為研究組(SCC),其中漢族40例,藏族38例,回族30例;收集同時期年齡相近的因子宮肌瘤在我院住院治療的107例正常宮頸組織標本為對照組(NCE),其中漢族40例,藏族37例,回族30例。免疫組化方法被用于檢測Merlin蛋白的表達情況;SPSS 20.0軟件被應用于進行數(shù)據(jù)處理分析,χ2檢驗和Fisher確實概率法驗證各組率的差異。結果:1.SCC組Merlin蛋白陽性表達率為17.6%(19/108),NCE組Merlin蛋白陽性表達率為89.7%(96/107),明顯比SCC組高(P0.05),二者對比差異顯著;其中NCE組40例漢族、37例藏族、30例回族Merlin蛋白陽性表達率分別為90.0%(36/40)、91.9%(34/37)、86.7%(26/30);SCC組40例漢族、38例藏族、30例回族Merlin蛋白陽性表達率分別為17.5%(7/40)、18.4%(7/38)、16.7%(5/30),在各種族中SCC組Merlin蛋白非陰性顯示結果比對NCE組都存在統(tǒng)計學差別(P0.05);2.漢、藏、回SCC中Merlin蛋白陽性表達結果經比對無民族差別(P0.05),同樣對比漢、藏、回NCE中Merlin蛋白陽性顯示結果亦未見民族差別(P0.05);3.SCC組中40例漢族,分別對Merlin蛋白在不同年齡、FIGO分期、淋巴結轉移情況及細胞分化程度中的陽性表達率進行了比較,未見在各臨床病理參數(shù)內Merlin蛋白非陰性顯示率存在明顯差別(P0.05),同樣對SCC組中38例藏族及30例回族中Merlin蛋白在各臨床病理參數(shù)中的陽性表達進行比較,均無明顯差異。結論:1.Merlin蛋白陽性顯示于青海女性子宮頸鱗癌中比非病變宮頸上皮顯著減少,提示子宮頸鱗癌的發(fā)生或許與Merlin蛋白顯示降低、缺失密切相關,Merlin蛋白檢測有助于輔助推斷子宮頸鱗癌發(fā)生病因;2.Merlin蛋白于漢、藏、回族非病變宮頸上皮及宮頸鱗狀細胞癌內顯示無民族差異,提示種族間子宮頸鱗癌發(fā)生率的差異可能不受Merlin蛋白表達的影響;3.漢、藏、回三民族中子宮頸鱗癌組織Merlin蛋白的表達均與患者年紀、FIGO分期、淋巴移動狀態(tài)、細胞分化水平等特性沒關系(P0.05),提示Merlin蛋白表達缺少或許只是子宮頸鱗癌形成的先期事件,并未直接參加子宮頸鱗癌的遠處移動、粘附擴散等流程。
[Abstract]:Objective: to explore the display status of NF2 gene Merlin in non-pathological cervical tissues and squamous cell carcinoma of cervix in Qinghai women, and to compare whether there are ethnic differences in the display of Merlin protein in cervical squamous cell carcinoma of Qinghai Han nationality, Tibetan nationality and Hui nationality. Furthermore, the display of Merlin protein in the stage of Figo, the status of lymphatic metastasis and the level of cell differentiation in patients with cervical squamous cell carcinoma of different nationalities were further discussed. Methods: from July 2014 to July 2016, 108 cases of cervical squamous cell carcinoma treated by radical cervical cancer in Qinghai Red Cross Hospital were collected as study group (40 cases of Han nationality, 38 cases of Tibetan nationality and 30 cases of Hui nationality). A total of 107 normal cervical tissue specimens of the same age, including 40 Han, 37 Tibetans and 30 Hui, were collected from normal cervix tissues for hysteromyoma in our hospital. Immunohistochemical method was used to detect the expression of Merlin protein. SPSS20.0 software was used for data processing and analysis. 蠂 2 test and Fisher positive probability method were used to verify the difference in the rates of each group. Results 1. The positive expression rate of Merlin protein in SCC group was 17.610 / 10 8% and the positive expression rate of Merlin protein was 89.7 / 96 / 107%, which was significantly higher than that in SCC group (P 0.05). The difference between the two groups was significant. The positive expression rate of Merlin protein in NCE group (40 cases), Han nationality (37 cases), Tibetan group (30 cases), Hui nationality (30 cases) was 90.0%, respectively. The positive expression rate of Merlin protein in SCC group was 17.5% 18.40% 18.440% 18.440% 18.440%, respectively, and the positive rate of Merlin protein in SCC group was 16.7% 30%. The non-negative expression rate of Merlin protein in SCC group was negative in all races. The positive expression rate of Merlin protein in 38 cases of Han nationality and 30 cases of Hui nationality was 17.55% -18.40% 18.44%, respectively. There was statistical difference in NCE group (P 0.05). The results of positive expression of Merlin protein in SCC of Han, Tibetan and Hui were compared with that of no ethnic difference (P0.05). Similarly, the positive results of Merlin protein in NCE of Han and Tibetan were also not found. 3. In the group of Han nationality, 40 cases of Han nationality in the group of NCE were divided into different age and stage of FIGO, respectively. The positive expression rates of lymph node metastasis and cell differentiation were compared. There was no significant difference in the non-negative display rate of Merlin protein among different clinicopathological parameters. There was no significant difference in the positive expression of Merlin protein in 38 cases of Tibetan nationality and 30 cases of Hui nationality in SCC group. Conclusion: 1. The presence of Merlin protein in cervical squamous cell carcinoma in Qinghai women was significantly lower than that in non-pathological cervical epithelium, suggesting that the occurrence of cervical squamous cell carcinoma might be lower than that of Merlin protein. Detection of Merlin protein is closely related to deletion. 2. There is no difference in the presence of Merlin protein in cervix epithelium and squamous cell carcinoma of cervix in Han, Zang and Hui nationality. The results suggest that the difference in incidence of cervical squamous cell carcinoma between ethnic groups may not be affected by the expression of Merlin protein. The expression of Merlin protein in cervical squamous cell carcinoma was correlated with Figo stage and lymphatic movement in Han, Zang and Hui Sanmin ethnic groups. It is suggested that the lack of expression of Merlin protein may be a preevent in the formation of squamous cell carcinoma of the cervix, and it does not directly participate in the process of distant migration, adhesion and diffusion of squamous cell carcinoma of the cervix.
