低表達(dá)長(zhǎng)非編碼RNA MALAT1對(duì)人宮頸癌HeLa細(xì)胞體外生長(zhǎng)和遷移的影響

【摘要】:設(shè)計(jì)3個(gè)針對(duì)MALAT1不同靶序列的小干擾RNA(small interfering RNA,siRNA),篩選出有效的siRNA序列,設(shè)計(jì)并構(gòu)建短發(fā)夾RNA(short hairpin RNA,shRNA)干擾質(zhì)粒,轉(zhuǎn)染到HeLa細(xì)胞中,構(gòu)建低表達(dá)MALAT1的穩(wěn)定細(xì)胞株.通過(guò)細(xì)胞的生長(zhǎng)曲線和細(xì)胞劃痕實(shí)驗(yàn)驗(yàn)證降低MALAT1對(duì)HeLa細(xì)胞增殖和遷移的影響.結(jié)果顯示MALAT1表達(dá)水平在HeLa細(xì)胞中得到有效地降低,低表達(dá)MALAT1的HeLa細(xì)胞生長(zhǎng)速度和遷移速度減小.表明MALAT1在HeLa細(xì)胞中具有調(diào)控細(xì)胞增殖和遷移的能力.
[Abstract]:Three small interfering RNA(small interfering siRNAs targeting different target sequences of MALAT1 were designed, and the effective siRNA sequences were screened out. The interference plasmids of short hairpin RNA(short hairpin RNAs were designed and constructed, and then transfected into HeLa cells to construct stable cell lines with low MALAT1 expression. The effect of MALAT1 on proliferation and migration of HeLa cells was confirmed by cell growth curve and cell scratch test. The results showed that the expression level of MALAT1 was significantly decreased in HeLa cells, and the growth and migration speed of HeLa cells with low MALAT1 expression was decreased. It is suggested that MALAT1 has the ability to regulate cell proliferation and migration in HeLa cells.
