Ang-1、Ang-2、TSP-1與復(fù)發(fā)性流產(chǎn)的相關(guān)性研究

【摘要】:目的 通過觀察復(fù)發(fā)性流產(chǎn)(RSA)患者絨毛、蛻膜組織中血管生成素-1(Ang-1)、血管生成素-2(Ang-2)、血小板反應(yīng)蛋白-1(TSP-1)的表達(dá)情況,探討Ang-1、Ang-2、TSP-1與RSA的關(guān)系,為臨床治療提供理論依據(jù)。 方法 選擇2013年5月--2013年11月在山西大醫(yī)院婦產(chǎn)科門診行早期人工流產(chǎn)的復(fù)發(fā)性流產(chǎn)患者30例為研究組,選擇同期自愿行人工流產(chǎn)、既往分娩過足月正常胎兒的正常早孕婦女30例為對(duì)照組,在人工流產(chǎn)的同時(shí)留取絨毛及蛻膜組織,經(jīng)免疫組化SP法檢測Ang-1、Ang-2及TSP-1在兩組絨毛及蛻膜組織中的表達(dá)水平,并對(duì)有關(guān)數(shù)據(jù)進(jìn)行統(tǒng)計(jì)分析。 結(jié)果 1. Ang-1、Ang-2、TSP-1在各組每例的絨毛及蛻膜中均有表達(dá)。研究組絨毛組織中Ang-1、Ang-2的表達(dá)高于對(duì)照組,TSP-1的表達(dá)低于對(duì)照組;蛻膜組織中Ang-1、Ang-2、TSP-1的表達(dá)均低于對(duì)照組,且差異均有統(tǒng)計(jì)學(xué)意義(P<0.05)。研究組絨毛Ang-1/Ang-2比值高于對(duì)照組,且差異有統(tǒng)計(jì)學(xué)意義(P<0.05);研究組蛻膜中Ang-1/Ang-2比值高于對(duì)照組,但無統(tǒng)計(jì)學(xué)意義(P0.05)。 2.研究組中,絨毛Ang-1、Ang-2及TSP-1表達(dá)均高于蛻膜,Ang-1/Ang-2比值低于蛻膜,且差異有統(tǒng)計(jì)學(xué)意義(P<0.05)。 3.對(duì)照組中,絨毛中Ang-1表達(dá)顯著低于蛻膜,TSP-1表達(dá)高于蛻膜,且差異有統(tǒng)計(jì)學(xué)意義(P<0.05),Ang-2在絨毛、蛻膜組織中表達(dá)無統(tǒng)計(jì)學(xué)差異(P0.05),絨毛Ang-1/Ang-2比值低于蛻膜,且有統(tǒng)計(jì)學(xué)差異(P<0.05)。 結(jié)論 1.Ang-1、Ang-2、TSP-1在反復(fù)自然流產(chǎn)患者絨毛及蛻膜組織的異常表達(dá)可能是造成復(fù)發(fā)性流產(chǎn)的原因。 2. Ang-1、Ang-2在蛻膜的低表達(dá)可能是導(dǎo)致其在絨毛中的高表達(dá)及Ang-1/Ang-2平衡失調(diào)的原因,,兩者共同作用導(dǎo)致局部血管生成障礙及流產(chǎn)的發(fā)生。 3. RSA患者絨毛組織中Ang-1過度表達(dá)對(duì)血管生成缺陷的影響可能更大。 4. TSP-1在絨毛及蛻膜中的低表達(dá)可能也是導(dǎo)致血管生成紊亂及RSA的原因。
[Abstract]:Purpose By observing the expression of Angiopoietin-1 (Ang-1), Ang-1 (Ang-1), Angiopoietin-2 (Ang-2) and platelet-reactive protein (-1) in the villi and decidua of patients with recurrent abortion, we investigated the relationship between Ang-1 and RSA, and provided a theoretical basis for clinical treatment. Method From May 2013 to November 2013, 30 patients with recurrent abortion who underwent early abortion in outpatient department of gynecology and obstetrics of Shanxi Great Hospital were selected as study group, and voluntary abortion was performed in the same period. 30 normal early pregnant women who had delivered term normal fetuses were used as control group. Villi and decidua tissues were taken simultaneously after induced abortion. The expression of Ang-1Ang-2 and TSP-1 in villi and decidua of the two groups were detected by immunohistochemical SP method. And carries on the statistical analysis to the related data. Result 1. Ang-1 and Ang-2 TSP-1 were expressed in villi and decidua of each group. The expression of Ang-1Ang-2 in chorionic villi in the study group was higher than that in the control group (P < 0.05), and that in decidua tissue was lower than that in the control group (P < 0.05). The ratio of Ang-1/Ang-2 in the study group was higher than that in the control group (P < 0.05) and the ratio of Ang-1/Ang-2 in decidua in the study group was higher than that in the control group (P < 0.05). 2. In the study group, the expression of Ang-1Ang-2 and TSP-1 in chorionic villi was higher than that in decidua Ang-1 / Ang-2 ratio, and the difference was statistically significant (P < 0.05). 3. In the control group, the expression of Ang-1 in chorionic villi was significantly lower than that in decidua, and the difference was statistically significant (P < 0.05). There was no significant difference in the expression of Ang-1/Ang-2 in chorionic villi. The Ang-1/Ang-2 ratio of villi was lower than that of decidua (P < 0.05). Conclusion 1. The abnormal expression of TSP-1 in villi and decidua of patients with recurrent spontaneous abortion may be the cause of recurrent abortion. 2. The low expression of Ang-1Ang-2 in decidua may be the cause of high expression of Ang-1Ang-2 in villi and imbalance of Ang-1/Ang-2 balance. 3. Overexpression of Ang-1 in chorionic villi of RSA patients may have a greater effect on angiogenesis defects. 4. The low expression of TSP-1 in chorionic villi and decidua may also be the cause of angiogenesis disorder and RSA.
