【摘要】：目的觀察Bax對人卵巢癌A2780細胞的凋亡誘導作用,并探討其作用機制。方法構建重組質粒pcDNA-Bax,將重組質粒pcDNA-Bax轉染A2780細胞,采用Hoechst 33258染色觀察細胞凋亡、MTT法檢測細胞活力,采用Western blot法檢測Bax基因的過表達、線粒體細胞色素C的釋放及Caspase-9和Caspase-3的激活。結果經酶切和測序鑒定,重組質粒pcDNA-Bax構建成功。轉染質粒pcDNA-Bax能明顯誘導A2780細胞凋亡,Hoechst染色顯示凋亡的細胞出現細胞核形態(tài)學改變;MTT結果顯示轉染質粒pcDNA3.1-Bax后,細胞活力明顯下降;Western blot結果表明轉染質粒pcDNA-Bax后的A2780細胞線粒體釋放細胞色素C,激活了Caspase-9和Caspase-3。結論 Bax能促進卵巢癌細胞線粒體細胞色素C釋放,從而誘導細胞凋亡。
[Abstract]:Objective to investigate the apoptosis-inducing effect of Bax on human ovarian cancer cell line A 2780 and its mechanism. Methods the recombinant plasmid pcDNA-Baxwas constructed. The recombinant plasmid pcDNA-Bax was transfected into A2780 cells. The viability of A2780 cells was detected by Hoechst 33258 staining. The overexpression of Bax gene, the release of mitochondrial cytochrome C and the activation of Caspase-9 and Caspase-3 were detected by Western blot assay. Results the recombinant plasmid pcDNA-Bax was successfully constructed by enzyme digestion and sequencing. Transfection of plasmid pcDNA-Bax could induce apoptosis of A2780 cells by Hoechst staining. The results of Western blot showed that the mitochondria of A2780 cells transfected with plasmid pcDNA-Bax released cytochrome C and activated Caspase-9 and Caspase-3. Conclusion Bax can promote the release of cytochrome C from mitochondria of ovarian cancer cells and induce apoptosis.
