PM2.5暴露對小鼠體外受精及胚胎發(fā)育的影響

【摘要】:目的研究PM2.5暴露對小鼠體外受精(IVF)及胚胎質(zhì)量的影響。方法選擇健康昆明雌鼠40只、雄鼠10只。40只雌小鼠隨機分成4組:PM2.5低劑量組(1.5 mg/kg)、PM2.5中劑量組(7.5 mg/kg)、PM2.5高劑量組(15 mg/kg)和對照組(給予相同體積的生理鹽水),每組10只。以氣管滴注的方式進行PM2.5染毒。比較各組間IVF基本情況(卵母細胞數(shù)、受精率、異常受精率、2PN卵裂率、2PN優(yōu)質(zhì)胚胎率、囊胚形成率)和卵泡液中相關(guān)細胞因子TNF-α和IFN-γ的表達。結(jié)果 PM2.5高劑量組的卵母細胞數(shù)、受精率、2PN卵裂率、2PN優(yōu)質(zhì)胚胎率、囊胚形成率與對照組相比降低,而異常受精率明顯增高,差異具有統(tǒng)計學(xué)意義(P0.05);PM2.5高劑量組卵泡液中相關(guān)細胞因子TNF-α和IFN-γ的表達明顯高于中劑量組與低劑量組,而中劑量組表達又高于低劑量組,組間比較差異具有統(tǒng)計學(xué)意義(P0.05),高、中、低劑量組表達均高于對照組,差異具有統(tǒng)計學(xué)意義(P0.05)。結(jié)論 PM2.5暴露會減少小鼠體外受精的獲卵數(shù);降低受精率、2PN卵裂率、2PN優(yōu)質(zhì)胚胎率、囊胚形成率。
[Abstract]:Objective to study the effects of PM2.5 exposure on in vitro fertilization (IVF) and embryo quality in mice. Methods Forty healthy Kunming female rats and 10 male mice were randomly divided into 4 groups: low dose group (1.5 mg / kg), middle dose group (7.5 mg / kg), high dose group (15 mg / kg) and control group (10 rats in each group) with the same volume of normal saline. PM2.5 was administered by trachea drip. The expression of cytokines TNF- 偽 and IFN- 緯 in follicular fluid were compared between groups (oocyte number, fertilization rate, abnormal fertilization rate and 2PN cleavage rate, blastocyst formation rate and blastocyst formation rate). Results the number of oocytes, fertilization rate and 2PN cleavage rate, blastocyst formation rate and abnormal fertilization rate in PM2.5 high dose group were significantly higher than those in control group. The expression of TNF- 偽 and IFN- 緯 in follicular fluid of high dose group was significantly higher than that of medium dose group and low dose group, while the expression of TNF- 偽 and IFN- 緯 in middle dose group was higher than that in low dose group. The expression of low dose group was higher than that of control group, and the difference was statistically significant (P 0.05). Conclusion PM2.5 exposure can reduce the number of eggs obtained by in vitro fertilization, reduce fertilization rate and 2PN cleavage rate, 2PN high quality embryo rate and blastocyst formation rate.
