【摘要】：目的檢測肝X受體蛋白在卵巢癌組織及正常卵巢組織中的表達,探討肝X受體激動劑T0901317對卵巢癌細胞系SKOV3細胞活力及凋亡的影響。方法收集2015年8月至2016年8月行初次手術(shù)治療的51例卵巢癌患者的組織標本,另選取32例因子宮肌瘤等良性病變切除的正常卵巢組織作對照。采用免疫組織化學(xué)方法檢測肝X受體蛋白在卵巢癌組織及正常卵巢組織中的表達;CCK8檢測T0901317對卵巢癌細胞系SKOV3增殖的影響;流式細胞術(shù)檢測T0901317對卵巢癌細胞系SKOV3凋亡的影響。結(jié)果肝X受體蛋白在正常卵巢組織較卵巢癌組織表達量高,兩組比較差異有統(tǒng)計學(xué)意義(P0.001),T0901317抑制卵巢癌細胞系SKOV3增殖具有明顯的劑量依賴性和時間依賴性(P0.05),T0901317促進SKOV3凋亡具有明顯的劑量依賴性(P0.05)。結(jié)論肝X受體表達下調(diào)可能與卵巢癌的發(fā)生發(fā)展有關(guān),激活肝X受體可能會成為卵巢癌潛在的治療靶點。
[Abstract]:Objective to detect the expression of liver X receptor protein in ovarian cancer and normal ovarian tissues, and to investigate the effects of T0901317, a hepatic X receptor agonist, on the viability and apoptosis of ovarian cancer cell line SKOV3. Methods from August 2015 to August 2016, the tissue specimens of 51 patients with ovarian cancer underwent initial surgical treatment were collected and 32 normal ovarian tissues were selected as control group, which were excised from benign lesions such as uterine leiomyoma. The expression of liver X receptor protein in ovarian cancer and normal ovarian tissues was detected by immunohistochemical method. The effect of T0901317 on the proliferation of ovarian cancer cell line SKOV3 and the effect of T0901317 on apoptosis of ovarian cancer cell line SKOV3 were detected by flow cytometry. Results the expression of X receptor protein in normal ovary was higher than that in ovarian cancer. There was a significant difference between the two groups in the inhibition of SKOV3 proliferation by P0.001T0901317 in a dose-dependent and time-dependent manner. P0.05T0901317 promoted SKOV3 apoptosis in a dose-dependent manner. Conclusion the down-regulation of liver X receptor expression may be related to the occurrence and development of ovarian cancer, and activation of liver X receptor may be a potential therapeutic target for ovarian cancer.
