性激素結合球蛋白、胰島素信號轉導蛋白和葡萄糖轉運蛋白在妊娠期糖尿病胎盤組織中的表達及相關性分析

【摘要】:目的研究妊娠期糖尿病(GDM)孕婦胎盤組織中性激素結合球蛋白(SHBG)、胰島素信號轉導蛋白及葡萄糖轉運蛋白表達,探討其在GDM發(fā)病過程中的作用。方法收集足月妊娠且非肥胖(BMI25 kg/m2)GDM孕婦(GDM組)和同期糖代謝正常(正常組)孕婦胎盤組織各10例,應用Western blotting、實時PCR方法檢測2組胎盤組織中SHBG和胰島素信號轉導蛋白(IRS-1、ISR-2、PI3K p85α)和葡萄糖轉運蛋白(GLUT-1、GLUT-3、GLUT-4)的表達,各指標進行直線回歸依存關系分析。結果與正常組比較,GDM組胎盤組織中SHBG m RNA、蛋白表達降低(P0.05);IRS-1蛋白、IRS-2 m RNA表達降低(P0.05);PI3K p85α和GLUT-1蛋白及其m RNA表達無統(tǒng)計學差異(P0.05);GLUT-3蛋白,GLUT-4 m RNA、蛋白表達降低(P0.05)。直線回歸分析結果顯示SHBG m RNA與IRS-2 m RNA、PI3K p85αm RNA、GLUT-4 m RNA的表達呈正相關(均P0.05);IRS-2 m RNA和GLUT-4 m RNA的表達呈正相關(P0.01);IRS-1 m RNA和PI3K p85αm RNA、GLUT-3 m RNA的表達呈負相關(P0.05);IRS-2m RNA和GLUT-1 m RNA的表達正相關(P0.01)。結論 GDM孕婦胎盤中胰島素信號轉導和葡萄糖轉運蛋白存在異常,SHBG可能參與胰島素信號通路的調節(jié),GDM時胎盤滋養(yǎng)細胞合成和分泌SHBG減少,引起胰島素信號轉導通路相關蛋白表達降低,從而導致胰島素抵抗及GDM的發(fā)生。
[Abstract]:Objective to study the expression of neutral hormone binding globulin (SHBG), insulin signal transduction protein and glucose transporter protein in pregnant women with gestational diabetes mellitus (GDM), and to explore its role in the pathogenesis of GDM. Methods the placenta of pregnant women of full-term pregnancy and non obese (BMI25 kg/m2) GDM pregnant women (group GDM) and normal group of glucose metabolism (normal group) were collected. The expression of SHBG and insulin signal transduction proteins (IRS-1, ISR-2, PI3K p85 a) and glucose transporters (GLUT-1, GLUT-3, GLUT-4) in the 2 groups of placental tissues were detected by Western blotting. The results were compared with the normal group. The results were compared with the normal group. The expression of white expression decreased (P0.05), and the expression of IRS-1 protein and IRS-2 m RNA decreased (P0.05); PI3K p85 A and GLUT-1 protein and m RNA expressed no statistical difference (P0.05). There is a positive correlation between the expression of IRS-2 m RNA and GLUT-4 m RNA (P0.01), IRS-1 m RNA and PI3K p85. At GDM, the synthesis of placental trophoblast cells and the secretion of SHBG decrease, resulting in a decrease in the expression of protein related proteins in the insulin signal transduction pathway, which leads to insulin resistance and the occurrence of GDM.
