母鼠營養(yǎng)不良導(dǎo)致子代在生命早期出現(xiàn)糖脂代謝紊亂及其機制探討

【摘要】:為了探討母鼠孕期和哺乳期營養(yǎng)不良對子代生命早期糖脂代謝的影響及其機制,文章對孕期和哺乳期母鼠分別喂養(yǎng)高脂飲食、低蛋白飲食和正常飲食,觀察其子鼠斷乳時(3周齡)糖脂代謝指標,并采用熒光定量PCR方法檢測子鼠肝組織氧化物酶增殖物激活受體γ(PPARγ)基因的表達情況。結(jié)果表明:子鼠在3周齡時,與正常飲食組相比,低蛋白飲食組子鼠出生體重(7.36±0.91 vs 8.94±1.39,P0.0001)較低,體長較短(12.27±0.53 vs13.44±0.36,P0.0001);高脂飲食組子鼠體重(9.53±0.68 vs 7.36±0.91,P0.0001)和體長(13.22±0.35 vs 12.27±0.53,P0.0001)均高于低蛋白飲食組;另外,高脂飲食組子鼠腹腔糖耐量實驗30 min和60 min血糖明顯高于正常飲食組(P0.001),且高脂飲食組30 min血糖水平也明顯高于低蛋白飲食組(P0.001),高脂飲食組子鼠糖耐量曲線下面積明顯大于正常飲食組(P0.001)。另外,與正常飲食組相比,高脂飲食組子鼠空腹膽固醇水平明顯升高(1.64±0.21 vs 1.18±0.16,P0.01),低蛋白飲食組空腹膽固醇水平明顯下降(0.96±0.09 vs 1.18±0.16,P0.05)。熒光定量PCR結(jié)果顯示,在低蛋白飲食組和高脂飲食組,其子鼠肝組織PPARγ基因表達量均明顯高于正常飲食組(P0.05)。結(jié)果顯示,母鼠妊娠期和哺乳期高脂飲食與低蛋白飲食均可以誘導(dǎo)子鼠在發(fā)育早期出現(xiàn)糖脂代謝紊亂,PPARγ基因可能在其中參與了重要的調(diào)控作用。
[Abstract]:In order to investigate the effect of maternal malnutrition during pregnancy and lactation on the metabolism of glucose and lipid in the early life of offspring, the high fat diet, low protein diet and normal diet were fed to pregnant and lactating mothers, respectively. The indexes of glucose and lipid metabolism were observed at the age of 3 weeks during weaning, and the expression of PPAR- 緯 gene in liver tissue was detected by fluorescence quantitative PCR. The results showed that compared with the normal diet group, the birth weight of the offspring in the low-protein diet group was 7.36 鹵0.91 vs 8.94 鹵1.39 P0.0001) and the body length was 12.27 鹵0.53 vs13.44 鹵0.36 P0.0001) in the low protein diet group, 9.53 鹵0.68 vs 7.36 鹵0.91g P0.0001 in the high-fat diet group, and 13.22 鹵0.35 vs 12.27 鹵0.53P0.0001 in the low protein diet group, and the body length was 13.22 鹵0.35 vs 12.27 鹵0.53P0.0001 in the high-fat diet group. The blood glucose levels in the high fat diet group were significantly higher than those in the normal diet group for 30 min and 60 min, and the blood glucose level in the high fat diet group at 30 min was significantly higher than that in the low protein diet group. The area under the glucose tolerance curve in the high fat diet group was significantly higher than that in the low protein diet group. It was significantly higher than that of normal diet group (P 0.001). In addition, compared with the normal diet group, the fasting cholesterol level in the high-fat diet group was significantly higher than that in the high-fat diet group (1.64 鹵0.21 vs 1.18 鹵0.16 P0.01g), and the fasting cholesterol level in the low-protein diet group was significantly lower than that in the low-protein diet group (0.96 鹵0.09 vs 1.18 鹵0.16 P0.05). The results of fluorescence quantitative PCR showed that the expression of PPAR 緯 gene was significantly higher in the liver tissues of the rats in the low protein diet group and the high fat diet group than that in the normal diet group (P 0.05). The results showed that both high fat diet and low protein diet during pregnancy and lactation could induce the disorder of glucose and lipid metabolism in the early stage of development. PPAR 緯 gene might play an important role in the regulation.
