【摘要】：目的:探討子癇前期患者外周靜脈血清中三磷酸腺苷結(jié)合盒轉(zhuǎn)運體A1(ABCA1)蛋白表達的特點,闡明ABCA1對內(nèi)皮細(xì)胞損傷的影響及可能的機制。方法:選取同期住院、年齡、孕周無明顯差異的子癇前期患者40例(按病情輕重分為輕度、重度組)及正常妊娠者20名,采用ELISA試劑盒定量檢測3組研究對象外周靜脈血清中ABCA1及氧化低密度脂蛋白(OX-LDL)表達水平的差異。原代分離并培養(yǎng)上述3組研究對象臍靜脈內(nèi)皮細(xì)胞(HUVECs),采用熒光標(biāo)記的牛血清白蛋白(BSA)檢測內(nèi)皮細(xì)胞的單層屏障功能;采用MTT法檢測內(nèi)皮細(xì)胞的增殖能力;采用硝酸還原酶法檢測內(nèi)皮細(xì)胞的一氧化氮(NO)合成能力;計算OX-LDL/ABCA1比值,并進行相關(guān)性分析。結(jié)果:與正常妊娠組比較,子癇前期組患者外周靜脈血清中ABCA1蛋白表達水平明顯降低(P0.05),而OX-LDL表達水平及OX-LDL/ABCA1比值明顯升高(P0.05);ABCA1與OX-LDL表達水平呈明顯負(fù)相關(guān)關(guān)系(r=-0.681,P0.05)。與正常妊娠組比較,子癇前期組患者通過單層內(nèi)皮細(xì)胞的BSA水平明顯升高(P0.05),內(nèi)皮細(xì)胞增殖能力明顯下降(P0.05);內(nèi)皮細(xì)胞分泌NO水平明顯降低(P0.05)。且病情越嚴(yán)重,上述改變越明顯。子癇前期組患者血清中OX-LDL/ABCA1比值與通過內(nèi)皮細(xì)胞的BSA水平呈正相關(guān)關(guān)系(r=0.791,P0.05),與內(nèi)皮細(xì)胞的增殖水平呈負(fù)相關(guān)關(guān)系(r=-0.812,P0.05),與內(nèi)皮細(xì)胞中NO水平也呈負(fù)相關(guān)關(guān)系(r=-0.635,P0.05)。結(jié)論:子癇前期患者血清中ABCA1蛋白表達水平明顯下降,引起氧化應(yīng)激,OX-LDL和ABCA1表達失衡,這與全身血管內(nèi)皮細(xì)胞的損傷密切相關(guān),可能是參與子癇前期發(fā)生發(fā)展的重要機制之一。
[Abstract]:Aim: to investigate the expression of adenosine triphosphate binding cassette transporter A1 (A1) ABCA1 protein in peripheral venous serum of patients with preeclampsia and to elucidate the effect of ABCA1 on endothelial cell injury and its possible mechanism. Methods: 40 patients with preeclampsia who were hospitalized, age and gestational week were divided into mild group and severe group according to the severity of the disease and 20 normal pregnant women. The expression of ABCA1 and oxidized low density lipoprotein (OX-LDL) in peripheral vein serum of the three groups were quantitatively detected by ELISA kit. The umbilical vein endothelial cells (HUVECs) were isolated and cultured in primary culture, the monolayer barrier function of endothelial cells was detected by fluorescence labeled bovine serum albumin (BSA), and the proliferation of endothelial cells was detected by MTT method. The nitric oxide (no) synthesis of endothelial cells was measured by nitrate reductase method, and the ratio of OX-LDL/ABCA1 was calculated and the correlation analysis was carried out. Results: compared with the normal pregnancy group, the expression of ABCA1 protein in the peripheral vein of preeclampsia group was significantly lower than that in the normal pregnancy group, while the expression of OX-LDL and the ratio of OX-LDL/ABCA1 were significantly increased in the preeclampsia group. There was a negative correlation between the expression level of ABCA1 and the expression level of OX-LDL in the preeclampsia group. Compared with the normal pregnancy group, the BSA level of the monolayer endothelial cells in the preeclampsia group was significantly higher than that in the normal pregnancy group, and the proliferation ability of the endothelial cells was decreased significantly (P 0.05), while the level of no secreted by the endothelial cells decreased significantly (P 0.05). The more serious the disease, the more obvious the above changes were. In preeclampsia group, the ratio of OX-LDL/ABCA1 in serum was positively correlated with the level of BSA passing through endothelial cells, negatively correlated with the level of proliferation of endothelial cells, and negatively correlated with the level of no in endothelial cells. Conclusion: the expression of ABCA1 protein in serum of patients with preeclampsia is obviously decreased, which leads to the imbalance of OX-LDL and ABCA1 expression in patients with preeclampsia, which is closely related to the injury of systemic vascular endothelial cells and may be one of the important mechanisms involved in the development of preeclampsia.
