【摘要】：目的:檢測(cè)腫瘤相關(guān)成纖維細(xì)胞Twist1基因表達(dá),探討腫瘤相關(guān)成纖維細(xì)胞高表達(dá)Twist1對(duì)卵巢癌侵襲能力的影響。方法:CSIOVDB數(shù)據(jù)庫檢索Twist1基因在卵巢各種組織類型中的表達(dá)水平。分離原代正常成纖維細(xì)胞(NOF),以及卵巢癌患者的原代腫瘤相關(guān)成纖維細(xì)胞(CAF),免疫熒光實(shí)驗(yàn)驗(yàn)證Twist1基因在NOF和卵巢癌CAF細(xì)胞中的表達(dá)差異。TCGA數(shù)據(jù)庫489例卵巢癌中從mRNA水平驗(yàn)證Twist1與CAF經(jīng)典活化指標(biāo)FAP的相關(guān)性,以及炎癥因子IL-6的相關(guān)性。GSEA(Gene Set Enrichment Analysis)軟件探究CAF高表達(dá)Twist1與IL-6通路富集的關(guān)系。Western blot法驗(yàn)證下調(diào)Twist1基因?qū)L-6表達(dá)的影響。Transwell實(shí)驗(yàn)探究卵巢癌CAF-腫瘤細(xì)胞共培養(yǎng)體系中IL-6對(duì)卵巢癌細(xì)胞系SKOV3侵襲能力的影響。結(jié)果:Twist1基因在卵巢癌CAF中的表達(dá)水平明顯高于腫瘤細(xì)胞和正常成纖維細(xì)胞。TCGA數(shù)據(jù)庫中Twist1基因和FAP及IL-6基因呈顯著正相關(guān)(P0.0001)。GSEA軟件分析結(jié)果示,卵巢癌患者中CAF高表達(dá)Twist1的同時(shí),腫瘤上皮表現(xiàn)為IL-6通路明顯富集(P0.05,FDR25%)。Western blot法結(jié)果表明,Twist1基因下調(diào)后,IL-6基因表達(dá)量明顯降低。在共培養(yǎng)體系中加IL-6中和抗體,卵巢癌細(xì)胞侵襲力明顯下降。結(jié)論:腫瘤相關(guān)成纖維細(xì)胞高表達(dá)Twist1,可能通過提高IL-6水平增強(qiáng)卵巢癌侵襲能力。
[Abstract]:Aim: to detect the expression of Twist1 gene in tumor-associated fibroblasts and to investigate the effect of overexpression of Twist1 on the invasiveness of ovarian cancer. Methods the expression level of Twist1 gene in ovarian tissues was searched by the: CSIOVDB database. Primary normal fibroblasts were isolated from normal fibroblasts and primary tumor-associated fibroblasts from patients with ovarian cancer were isolated. Immunofluorescence assay verified the difference of expression of Twist1 gene in NOF and ovarian cancer CAF cells. TCGA database from 489 cases of ovarian cancer The correlation between Twist1 and FAP, a classical activation index of CAF, was verified. Correlation of inflammatory Factor IL-6 .GSEA Gene Set Enrichment Analysis) Software to explore the relationship between CAF overexpression of Twist1 and enrichment of IL-6 Pathway. Western blot method to verify the effect of down-regulation of Twist1 gene on IL-6 expression .Transwell experiment to explore IL-6 in ovarian cancer CAF- tumor cell coculture system Effect on invasion ability of ovarian cancer cell line SKOV3. Results the expression level of Twist1 gene in ovarian cancer CAF was significantly higher than that in tumor cell and normal fibroblast. TCGA database showed that there was a significant positive correlation between Twist1 gene, FAP gene and IL-6 gene. GSEA software analysis showed that CAF overexpression of Twist1 was also found in ovarian cancer patients. The expression of IL-6 gene was significantly decreased after down-regulation of Twist1 gene. The invasiveness of ovarian cancer cells decreased significantly with the addition of IL-6 neutralizing antibody in co-culture system. Conclusion: Twist1 is highly expressed in tumor-associated fibroblasts, which may enhance the invasiveness of ovarian cancer by increasing the level of IL-6.
