黃體酮和LPS-RS對流產(chǎn)大鼠的作用及大鼠蛻膜中IL-1β、TNF-α的表達(dá)

【摘要】:目的:通過尾靜脈注射脂多糖(LPS)建立大鼠流產(chǎn)的模型,用黃體酮及脂多糖拮抗劑LPS-RS干預(yù),計(jì)算胚胎吸收率,并檢測流產(chǎn)大鼠蛻膜組織中IL-1β、TNF-α因子蛋白表達(dá)情況,從而探討感染性流產(chǎn)的發(fā)病機(jī)制及黃體酮和LPS-RS對感染引起流產(chǎn)的作用。 方法:將雌雄大鼠合籠,發(fā)現(xiàn)陰栓日為妊娠第0天,孕鼠隨機(jī)分組,建立五組大鼠模型,實(shí)驗(yàn)一組為對照組,于孕7天尾靜脈注射磷酸緩沖液(PBS)1ml/只,實(shí)驗(yàn)二組于孕7天尾靜脈注射LPS1ml/只,實(shí)驗(yàn)三組于孕6-11天黃體酮肌注0.2ml/只,孕7天尾靜脈注射LPS1ml/只,實(shí)驗(yàn)四組于孕7天尾靜脈注射LPS-RS1ml/只后1小時再次尾靜脈注射LPS1ml/只,實(shí)驗(yàn)五組于孕6-11天加黃體酮肌注0.2ml/只,孕7天尾靜脈注射LPS-RS1ml/只后1小時再次尾靜脈注射LPS1ml/只,分組即為對照組、LPS組(流產(chǎn)組)、LPS+黃體酮組,LPS+LPS-RS組,LPS+黃體酮+LPS-RS組,并于孕13天處死,取流產(chǎn)大鼠的蛻膜組織,計(jì)算胚胎吸收率,比較各組之間的差異,用western blot方法檢測大鼠蛻膜組織IL-1β、TNF-α蛋白表達(dá)情況。 結(jié)果:五組胚胎吸收率分別為6.58%、24.79%、16.30%。9.89%、13.48%,實(shí)驗(yàn)二組(流產(chǎn)組)胚胎吸收率明顯高于實(shí)驗(yàn)一組(對照組),成功建立大鼠流產(chǎn)模型,實(shí)驗(yàn)三組、實(shí)驗(yàn)四組、實(shí)驗(yàn)五組與實(shí)驗(yàn)一組比較,胚胎吸收率均高于實(shí)驗(yàn)一組,但無統(tǒng)計(jì)學(xué)意義,實(shí)驗(yàn)四組、實(shí)驗(yàn)五組與實(shí)驗(yàn)二組比較,胚胎吸收率均低于實(shí)驗(yàn)二組,有統(tǒng)計(jì)學(xué)意義,實(shí)驗(yàn)三組、實(shí)驗(yàn)四組、實(shí)驗(yàn)五組胚胎吸收率情況,兩兩比較無明顯差異,無統(tǒng)計(jì)學(xué)意義。Western blot方法檢測大鼠蛻膜組織的IL-1p蛋白表達(dá)情況的比較:實(shí)驗(yàn)一組、實(shí)驗(yàn)五組與實(shí)驗(yàn)二組(流產(chǎn)組)比較,實(shí)驗(yàn)二組大鼠蛻膜組織IL-1p表達(dá)上升,P0.05,有統(tǒng)計(jì)學(xué)意義,實(shí)驗(yàn)三組、實(shí)驗(yàn)四組、實(shí)驗(yàn)五組間兩兩比較大鼠蛻膜組織IL-1p表達(dá)未見明顯差異,P0.05,無統(tǒng)計(jì)學(xué)意義。Western blot方法檢測大鼠蛻膜組織的TNF-α表達(dá)情況:實(shí)驗(yàn)一組、實(shí)驗(yàn)五組與實(shí)驗(yàn)二組(流產(chǎn)組)比較,實(shí)驗(yàn)二組大鼠蛻膜組織TNF-α表達(dá)上升,P0.05,有統(tǒng)計(jì)學(xué)意義,實(shí)驗(yàn)三組、實(shí)驗(yàn)四組、實(shí)驗(yàn)五組間兩兩比較大鼠蛻膜組織TNF-α表達(dá)未見明顯差異,P0.05,無統(tǒng)計(jì)學(xué)意義。 結(jié)論:脂多糖可引起妊娠大鼠流產(chǎn),用LPS-RS及黃體酮處理后能一定程度上拮抗LPS導(dǎo)致的胚胎吸收。流產(chǎn)大鼠蛻膜組織中IL-1p、TNF-α蛋白表達(dá)增加,而黃體酮與LPS-RS合用后能使這兩種細(xì)胞因子表達(dá)下調(diào),提示激素合并抗炎治療感染性流產(chǎn)效果更好。
[Abstract]:Objective: to establish a rat model of abortion by injecting lipopolysaccharide (LPS) into caudal vein. The embryo absorption rate was calculated with progesterone and lipopolysaccharide antagonist LPS-RS, and the expression of IL-1 尾 TNF- 偽 protein in decidua of aborted rats was detected. To explore the pathogenesis of infective abortion and the effect of progesterone and LPS-RS on infective abortion. Methods: the female and male rats were caged, and it was found that the day of Yin suppository was the first day of pregnancy. The pregnant rats were randomly divided into five groups. The experimental group was used as the control group. The rats were injected with phosphate buffer solution (PBS1ml / rat) at the end of the 7th day of gestation. LPS1ml/ was injected into the caudal vein on the 7th day of gestation, 0.2ml/ was injected intramuscularly by progesterone on the 6th to 11th day of gestation, LPS1ml/ was injected into the caudal vein on the 7th day of pregnancy, and LPS1ml/ was injected again one hour after LPS-RS1ml/ was injected into the caudal vein on the 7th day of gestation. The experimental five groups received intramuscular injection of 0.2ml/ with progesterone on 6-11 days of gestation, followed by re-injection of LPS1ml/ 1 hour after LPS-RS1ml/ was injected into the caudal vein on the 7th day of gestation. The rats were divided into two groups: control group (abortion group) and LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-LPS-RS group. On the 13th day of gestation, the decidua tissues of aborted rats were taken, the embryonic absorption rate was calculated, and the differences among the groups were compared. The expression of IL-1 尾 -TNF- 偽 protein in decidua tissue of rats was detected by western blot method. Results: the embryo absorption rates of the five groups were 6.58 and 24.79 respectively. The embryo absorption rate of experimental group 2 (abortion group) was significantly higher than that of experimental group 1 (control group, successful establishment of rat abortion model, experimental group 3, experimental group 4, experimental group 5 and experimental group 1), and the embryo absorption rate of experimental group 2 (abortion group) was significantly higher than that of experimental group 1 (control group, experiment 3, experiment 4, experiment 5 and experiment 1). The embryo absorptivity of experiment group was higher than that of experimental group 1, but there was no statistical significance. Compared with experimental group 2, the embryo absorption rate of experimental group 4 and experimental group 5 was lower than that of experimental group 2, which had statistical significance. There was no significant difference in embryo absorption rate among the five experimental groups. There was no statistical significance. Western blot method was used to detect the expression of IL-1p protein in decidua of rats: group 1, group 5 and group 2 (abortion group) were compared with each other. The expression of IL-1p in decidual tissue of experimental group 2 was significantly higher than that of control group (P 0.05). There was no significant difference in the expression of IL-1p in decidua tissues between two groups. There was no statistical significance. Western blot method was used to detect the expression of TNF- 偽 in decidual tissues of rats: compared with experimental group 1, experimental group 5 and experimental group 2 (abortion group), the expression of TNF- 偽 in decidual tissue of rats was detected by Western blot. The expression of TNF- 偽 in decidua of experimental group 2 was significantly higher than that of control group (P 0.05). There was no significant difference in expression of TNF- 偽 between experimental group 3, experimental group 4 and experimental group 5 (P 0.05). Conclusion: lipopolysaccharide can induce abortion in pregnant rats. LPS-RS and progesterone can antagonize embryo absorption induced by LPS to some extent. The expression of IL-1pnTNF- 偽 protein in decidual tissue of abortive rats was increased, but progesterone combined with LPS-RS could down-regulate the expression of these two cytokines, suggesting that hormone combined with anti-inflammatory therapy is better in the treatment of infective abortion.
