卵丘顆粒細胞SIRT1表達對卵胞漿內(nèi)單精子顯微注射助孕結(jié)局的影響

【摘要】:目的探討卵丘顆粒細胞SIRT1表達對ICSI助孕結(jié)局的影響。方法收集120例因男方因素在我院輔助生殖技術(shù)研究室行ICSI助孕的女性患者卵丘顆粒細胞,采用免疫組化法檢測SIRT1蛋白。比較分析ICSI妊娠與非妊娠組SIRT1表達率,并分析其與ICSI實驗室各項指標如穿卵泡數(shù)、獲卵數(shù)、獲卵率,受精數(shù)、受精率,卵裂數(shù)、卵裂率,優(yōu)質(zhì)胚胎數(shù)、囊胚形成數(shù)及囊胚冷凍數(shù)之間關(guān)系。結(jié)果妊娠與未妊娠組SIRT1表達率差異有統(tǒng)計學意義(P0.05)。SIRT1表達率與穿卵泡數(shù)、Ⅲ級OCCs數(shù)、ICSI卵子數(shù)、受精數(shù)、卵裂數(shù)、平均優(yōu)質(zhì)胚胎數(shù)、平均囊胚數(shù)、優(yōu)質(zhì)囊胚數(shù)、平均冷凍胚胎數(shù)間均有顯著相關(guān)性(P0.01)。結(jié)論 SIRT1在卵丘顆粒細胞表達可能影響卵母細胞成熟及卵子發(fā)育能力,進而與ICSI助孕結(jié)局有關(guān)。
[Abstract]:Objective to investigate the effect of SIRT1 expression in cumulus granulosa cells on the outcome of ICSI assisted pregnancy. Methods the cumulus granulosa cells were collected from 120 female women who were pregnant with ICSI because of male factors in the assisted reproductive technology laboratory of our hospital. The SIRT1 protein was detected by immunohistochemical method. The expression rate of SIRT1 in ICSI pregnancy group and non-pregnancy group was compared and compared with ICSI laboratory indexes such as the number of follicles pierced, the rate of obtaining eggs, the number of fertilization, the rate of fertilization, the number of cleavage, the rate of cleavage and the number of high-quality embryos. The relationship between the number of blastocysts and the number of frozen blastocysts. Results there were significant differences in SIRT1 expression rate between pregnant and non-pregnant women (P 0.05). SIRT1 expression rate was significantly higher than that in unpregnant women (P 0.05). The number of follicles, the number of SIRT1 eggs, fertilization, cleavage, average number of high quality embryos, average number of blastocysts, number of high quality blastocysts, and the number of high quality blastocysts were significantly different between pregnant and non-pregnant women. There was a significant correlation between the average number of frozen embryos and that of P0.01. Conclusion the expression of SIRT1 in cumulus granulosa cells may affect oocyte maturation and oocyte development, which may be related to the outcome of ICSI assisted pregnancy.
