【摘要】：目的:檢測(cè)miR23b和特異蛋白1(specific protein 1,Sp1)在卵巢子宮內(nèi)膜異位癥中的表達(dá)和分布情況,探討二者在卵巢子宮內(nèi)膜異位癥發(fā)生發(fā)展中的作用及意義。方法:qPCR檢測(cè)miR23b和Sp1 mRNA在異位內(nèi)膜、在位內(nèi)膜、正常內(nèi)膜中的表達(dá)水平;免疫組織化學(xué)和Western印跡檢測(cè)Sp1蛋白在異位內(nèi)膜、在位內(nèi)膜、正常內(nèi)膜的表達(dá)和分布情況。結(jié)果:MiR23b mRNA在異位內(nèi)膜、在位內(nèi)膜、正常內(nèi)膜組中的表達(dá)水平依次增加(P0.05)。Sp1在子宮內(nèi)膜間質(zhì)細(xì)胞、腺上皮細(xì)胞中均有表達(dá),主要表達(dá)在細(xì)胞核,少量表達(dá)在胞質(zhì)中;Sp1 mRNA和蛋白在異位內(nèi)膜、在位內(nèi)膜、正常內(nèi)膜中的表達(dá)均依次降低(P0.05)。MiR23b與Sp1 mRNA表達(dá)呈負(fù)相關(guān)(r= 0.526,P0.05)。結(jié)論:MiR23b和Sp1與卵巢子宮內(nèi)膜異位癥的發(fā)生發(fā)展密切相關(guān),二者可能通過(guò)相互拮抗,共同參與異位病灶的形成。
[Abstract]:Objective: to detect the expression and distribution of miR23b and specific protein 1(specific protein 1 / Sp1 in ovarian endometriosis and to explore the role and significance of them in the occurrence and development of ovarian endometriosis. Methods the expression and distribution of miR23b and Sp1 mRNA in ectopic, eutopic and normal endometrium were detected by 1: qPCR, and the expression and distribution of Sp1 protein in ectopic, eutopic and normal endometrium were detected by immunohistochemistry and Western blot. Results the expression of mRNA in ectopic endometrium, eutopic endometrium, normal endometrium and normal endometrium increased in turn, and the expression of P0.05U 路Sp1 in stromal cells and glandular epithelial cells of endometrium, mainly in the nucleus, was observed in both endometrium and glandular epithelium. The expression of Sp1 mRNA and protein in ectopic endometrium, eutopic endometrium and normal endometrium decreased in turn, and the expression of P0.05N. MiR23b was negatively correlated with Sp1 mRNA expression. Conclusion the ratio MiR23b and Sp1 are closely related to the occurrence and development of ovarian endometriosis. They may be involved in the formation of ectopic lesions through mutual antagonism.
