【摘要】：目的：探討microRNA-138、microRNA-720在新疆南部維吾爾族HPV16相關(guān)宮頸鱗癌組織中表達(dá)的差異。方法：采用瓊脂糖凝膠電泳技術(shù)檢測(cè)4例宮頸癌細(xì)胞系、83例新疆南部維吾爾族宮頸鱗癌組織中HPV16的表達(dá),采用實(shí)時(shí)定量RT-PCR技術(shù)對(duì)4例宮頸癌細(xì)胞系、83例新疆南部維吾爾族宮頸鱗癌組織標(biāo)本、16例與之對(duì)應(yīng)的癌旁組織標(biāo)本及40例新疆南部維吾爾族正常宮頸組織標(biāo)本檢測(cè)目的基因microRNA-138、microRNA-720的表達(dá)。結(jié)果：與癌旁組相比,microRNA-138、 microRNA-720在新疆南部維吾爾族宮頸鱗癌組中的表達(dá)均下調(diào),差異有統(tǒng)計(jì)學(xué)意義(P0.05)；與正常宮頸組相比,microRNA-138、microRNA-720在新疆南部維吾爾族宮頸鱗癌組中的表達(dá)均下調(diào),差異有統(tǒng)計(jì)學(xué)意義(P0.05)；與HPV16陰性宮頸鱗癌組相比,microRNA-138、microRNA-720在HPV16陽(yáng)性新疆南部維吾爾族宮頸鱗癌組中的表達(dá)均有下調(diào)趨勢(shì),但差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)；與HPV16陰性宮頸癌細(xì)胞系相比,microRNA-138、microRNA-720在HPV16陽(yáng)性宮頸癌細(xì)胞系中的表達(dá)無(wú)明顯差異(P0.05)。而且,下調(diào)的miRNA-138、miRNA-720均促進(jìn)淋巴結(jié)轉(zhuǎn)移、脈管浸潤(rùn)(P0.05),但與年齡、宮頸壁累及范圍無(wú)明顯關(guān)系(P0.05)；miRNA-720與臨床分期、腫瘤體積有關(guān)(P0.05)；但miRNA-138與臨床分期、腫瘤體積無(wú)明顯相關(guān)性(P0.05)。結(jié)論：MiRNA-138、miRNA-720與新疆南部維吾爾族宮頸鱗癌密切相關(guān),并與淋巴結(jié)轉(zhuǎn)移、脈管浸潤(rùn)有關(guān),但與HPV16無(wú)明顯相關(guān)性。miRNA-720與臨床分期、腫瘤體積也具有相關(guān)性。
[Abstract]:Objective: to investigate the expression of microRNA-138C microRNA-720 in Uygur cervix squamous cell carcinoma (SCC) tissues of Uygur nationality in southern Xinjiang. Methods: the expression of HPV16 was detected by agarose gel electrophoresis in 83 cases of cervical squamous cell carcinoma of Uygur nationality in southern Xinjiang. Real-time quantitative RT-PCR technique was used to detect 4 cervical cancer cell lines and 83 specimens of cervical squamous cell carcinoma tissues from Uygur nationality in southern Xinjiang. 16 specimens from adjacent tissues and 40 specimens from normal tissues of Uygur nationality in southern Xinjiang were detected. Objective to express the microRNA-720 gene. Results: compared with the paracancerous group, the expression of microRNA-720 was down-regulated in Uygur cervix squamous cell carcinoma group in the south of Xinjiang, the difference was statistically significant (P 0.05), and the expression of microRNA-138 microRNA-720 was down-regulated in Uygur cervix squamous cell carcinoma group in the south of Xinjiang compared with the normal cervix group. The expression of microRNA-138 microRNA-720 was down-regulated in HPV16 positive Uygur cervix cervix squamous cell carcinoma group in southern Xinjiang. However, there was no significant difference in the expression of microRNA-138 microRNA-720 in HPV16 positive cervical cancer cell line compared with HPV16 negative cervical cancer cell line. Moreover, the down-regulated miRNA-138miRNA-720 promoted lymph node metastasis and vessel invasion (P0.05), but had no significant relationship with age, cervical wall involvement and clinical stage, tumor volume was related to P0.05, but miRNA-138 had no significant correlation with clinical stage. ConclusionMiRNA-138miRNA-720 is closely related to squamous cell carcinoma of cervix of Uygur cervix in southern Xinjiang, and is related to lymph node metastasis and vascular invasion, but not to HPV16. MiRNA-720 is correlated with clinical stage and tumor volume.
