β-catenin在卵巢子宮內(nèi)膜異位癥在位及異位內(nèi)膜間質(zhì)細(xì)胞的表達(dá)及意義

【摘要】:目的: 體外分離培養(yǎng)卵巢子宮內(nèi)膜異位癥患者在位內(nèi)膜及異位內(nèi)膜間質(zhì)細(xì)胞,并探討-catenin在卵巢子宮內(nèi)膜異位癥在位及異位子宮內(nèi)膜間質(zhì)細(xì)胞的表達(dá)及其意義。 方法: 使用膠原酶消化聯(lián)合二次篩網(wǎng)過(guò)濾法體外分離、純化及培養(yǎng)卵巢子宮內(nèi)膜異位癥患者在位、異位和對(duì)照組子宮內(nèi)膜間質(zhì)細(xì)胞,分別應(yīng)用逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(RT-PCR)、Wester-blot方法檢測(cè)上述各組細(xì)胞中-catenin的mRNA和蛋白表達(dá)情況。 結(jié)果: 分離得到的內(nèi)膜間質(zhì)細(xì)胞純度達(dá)95%。OEms在位內(nèi)膜、異位內(nèi)膜和對(duì)照組中-catenin mRNA的IOD值分別為0.672±0.07,0.351±0.029和0.184±0.020,各組間均有統(tǒng)計(jì)學(xué)差異(F=27.68,P<0.001)。OEms在位內(nèi)膜、異位內(nèi)膜和對(duì)照組中-catenin蛋白的IOD值分別為0.803±0.074,,0.388±0.044和0.186±0.026,各組間均有統(tǒng)計(jì)學(xué)差異(F=36.83,P<0.001)。-catenin mRNA和蛋白的表達(dá)均在OEms在位內(nèi)膜最高,異位內(nèi)膜次之。 結(jié)論: 成功體外分離培養(yǎng)原代子宮內(nèi)膜間質(zhì)細(xì)胞,鑒定其純度達(dá)95%,可以為EMs的研究提供理想模型。OEMs在位、異位內(nèi)膜及正常對(duì)照組中均有β-cateninmRNA和蛋白的表達(dá);β-catenin在在位內(nèi)膜和異位內(nèi)膜的表達(dá)明顯高于正常對(duì)照組,其中在位內(nèi)膜表達(dá)最高。-catenin分子可能在EMs在位內(nèi)膜生物學(xué)改變及異位內(nèi)膜完成“三部曲”(即黏附、侵襲和血管形成)中起著重要作用,與EMs的發(fā)生發(fā)展密切相關(guān)。
[Abstract]:Objective: The eutopic endometrium and ectopic endometrial stromal cells were isolated from patients with ovarian endometriosis in vitro. The expression and significance of -catenin in eutopic and ectopic endometrial stromal cells of patients with ovarian endometriosis were investigated. Methods: The eutopic, ectopic and control endometrial stromal cells of patients with ovarian endometriosis were purified and cultured by collagenase digestion combined with secondary screen filtration in vitro. The mRNA and protein expression of -catenin were detected by reverse transcriptase polymerase chain reaction (RT-PCR) and Western blot. Results: The purity of the isolated endometrial stromal cells was as high as that of 95%.OEms eutopic endometrium. The IOD values of -catenin mRNA in ectopic endometrium and control group were 0.672 鹵0.07 鹵0.351 鹵0.029 and 0.184 鹵0.020, respectively. There was significant difference between the two groups (P < 0.001).OEms). The IOD values of -catenin protein in ectopic endometrium and control group were 0.803 鹵0.074 鹵0.388 鹵0.044 and 0.186 鹵0.026, respectively. There was a significant difference in the expression of -catenin mRNA and protein between ectopic endometrium and ectopic endometrium. The expression of -catenin mRNA and protein in ectopic endometrium was the highest in OEms eutopic endometrium, followed by ectopic endometrium. Conclusion: The primary endometrial stromal cells were successfully isolated and cultured in vitro, and their purity was 95%, which could provide an ideal model for the study of EMs. The expression of 尾 -catenin mRNA and protein in ectopic endometrium and normal control group was significantly higher than that in eutopic endometrium and ectopic endometrium, and the expression of 尾 -catenin in eutopic endometrium and ectopic endometrium was significantly higher than that in normal control group. The highest expression of .-catenin in eutopic endometrium may play an important role in the biological changes of eutopic endometrium and the completion of "trilogy" (adhesion, invasion and angiogenesis) in eutopic endometrium of EMs, which is closely related to the occurrence and development of EMs.
