PLGA生物可降解膜用于預(yù)防宮腔粘連的初步研究

【摘要】:目的:研究聚乳酸-羥基乙酸共聚物[poly(lactic-co-glycolic acid),PLGA]膜作為生物可降解材料用于預(yù)防宮腔粘連的可行性初步研究。方法:制備不同單體比例的PLGA膜,分別于模擬宮腔液以及昆明小鼠宮腔中進(jìn)行體外和體內(nèi)降解實(shí)驗(yàn)。將30只昆明小鼠隨機(jī)分為假手術(shù)組、宮腔粘連組、PLGA膜組,每組各10只,運(yùn)用Masson染色和Western blot觀察各組昆明小鼠子宮內(nèi)膜纖維化程度和相關(guān)蛋白的表達(dá)。結(jié)果:不同單體比例的PLGA膜體外降解時(shí)間不同,PLGA膜(50∶50)的降解速度最快,在4周時(shí)質(zhì)量損失達(dá)98.5%;PLGA膜(90∶10)降解速度最慢,在12周時(shí)其質(zhì)量損失僅為7.8%;PLGA(75∶25)降解速度介于前兩者之間,于12周時(shí)質(zhì)量損失達(dá)95.7%。不同單體比例的PLGA膜體內(nèi)降解時(shí)間不同,PLGA膜(50∶50)7 d后在宮腔內(nèi)完全降解,而PLGA膜(75∶25)、PLGA膜(90∶10)仍然滯留于宮腔中。與假手術(shù)組相比,PLGA膜組(75∶25)增加了昆明小鼠子宮內(nèi)膜纖維化面積(P=0.000)以及相關(guān)蛋白的表達(dá)(P0.01);與宮腔粘連組相比,PLGA膜組(75∶25)子宮內(nèi)膜纖維化面積以及相關(guān)蛋白的表達(dá)無明顯差異(P0.05)。結(jié)論:PLGA生物可降解材料用于預(yù)防宮腔粘連,具有宮腔內(nèi)降解速度可調(diào)節(jié)、不增加宮腔粘連組子宮內(nèi)膜纖維化程度的特點(diǎn),在預(yù)防宮腔粘連方面具有潛在的應(yīng)用價(jià)值。
[Abstract]:Aim: to study the feasibility of poly(lactic-co-glycolic acidoglycolic acid (poly(lactic-co-glycolic) membrane as biodegradable material for preventing intrauterine adhesion. Methods: PLGA membranes with different monomer ratios were prepared and degraded in vitro and in vivo in simulated uterine fluid and Kunming mouse uterine cavity respectively. Thirty Kunming mice were randomly divided into sham operation group and intrauterine adhesion group with 10 mice in each group. The degree of endometrial fibrosis and the expression of related proteins in Kunming mice were observed by Masson staining and Western blot. Results: the degradation rate of PLGA membrane with different monomer ratio in vitro was the fastest, and the mass loss reached 98.55% at 4 weeks. The degradation rate of PLGA 75: 25 was between the former two at 12 weeks, and the mass loss reached 95.7at 12 weeks. The degradation time of PLGA membrane with different monomer ratio was 50: 507 days later, and PLGA membrane 75: 25 / PLGA membrane 90: 10 remained in uterine cavity. Compared with sham operation group, PLGA membrane group (75: 25) increased the area of endometrial fibrosis in Kunming mice (P0. 000) and the expression of related protein (P0. 01); compared with the group of intrauterine adhesion, PLGA membrane group (75: 25) increased the area of endometrial fibrosis and related proteins. There was no significant difference in expression of P0.05. Conclusion the biodegradable material of% PLGA can be used to prevent intrauterine adhesion, which has the characteristics of adjustable degradation rate in uterine cavity and no increase in the degree of endometrial fibrosis in the group of intrauterine adhesions. It has potential application value in the prevention of intrauterine adhesions.
