【摘要】：目的探討調(diào)節(jié)性T細(xì)胞(Treg)和Notch1信號(hào)通路在原因不明復(fù)發(fā)性自然流產(chǎn)(URSA)中的作用。方法流式細(xì)胞儀檢測(cè)URSA患者(URSA組)及正常妊娠婦女(對(duì)照組)蛻膜CD4~+CD25~+T細(xì)胞Treg表達(dá)比例,real time RT-PCR及Western blotting檢測(cè)蛻膜中CD4~+T細(xì)胞中Notch1信號(hào)通路和叉頭轉(zhuǎn)錄因子家族3(Foxp3)表達(dá)情況。結(jié)果 URSA組CD4~+CD25~+T細(xì)胞/淋巴細(xì)胞、CD4~+Foxp3~+T細(xì)胞/淋巴細(xì)胞和CD4~+Foxp3~+T細(xì)胞/CD4~+T細(xì)胞比例均低于對(duì)照組(P0.05)。URSA組CD4~+T細(xì)胞中Notch1-Ic、RBPJκ、Foxp3 m RNA及蛋白表達(dá)均低于對(duì)照組。結(jié)論 URSA患者蛻膜CD4~+T細(xì)胞中Notch1信號(hào)通路和Foxp3表達(dá)下調(diào),CD4~+CD25~+T細(xì)胞表達(dá)比例下降,提示URSA患者Notch1信號(hào)通路和Foxp3表達(dá)下調(diào)可能阻礙CD4~+T細(xì)胞轉(zhuǎn)化為CD4~+CD25~+T細(xì)胞,進(jìn)而誘發(fā)免疫排斥,誘導(dǎo)流產(chǎn)。
[Abstract]:Objective to investigate the regulatory T cells (Treg) and Notch1 signaling pathway in unexplained recurrent spontaneous abortion (URSA) in vitro. Methods flow cytometry was used to detect URSA patients (URSA group) and normal pregnant women (control group) decidual CD4~+CD25~+T cells Treg expression ratio of real time RT-PCR Western blotting, and CD4~+T in decidual detection cells in the Notch1 signaling pathway and the forkhead transcription factor family 3 (Foxp3) expression. Results CD4~+CD25~+T cells / URSA group CD4~+Foxp3~+T cells / lymphocytes, percentage of lymphocytes and CD4~+Foxp3~+T cells in /CD4~+T cells were lower than the control group (P0.05) Notch1-Ic,.URSA group CD4~+T cells RBPJ K, m RNA and protein expression of Foxp3 was lower than the control group. The down-regulation of Notch1 signaling pathway and the expression of Foxp3 in patients with URSA conclusion decidual CD4~+T cells, CD4~+CD25~+T cells decreased the proportion of patients with Notch1 suggest that URSA signaling pathway and Foxp3 Down regulation may inhibit the transformation of CD4~+T cells into CD4~+CD25~+T cells, and then induce immune rejection and induce abortion.
