【摘要】：目的：探究脾酪氨酸激酶(spleen tyrosine kinase, Syk)及促凋亡線粒體蛋白（Bcl-2adenocarcinoma E1B19kD interacting protein3，BNip3）在子宮內(nèi)膜癌組織、子宮內(nèi)膜不典型增生組織及其正常子宮內(nèi)膜組織中的表達及其相關(guān)臨床意義。 方法：采用實時定量熒光PCR和免疫組化的方法測定SYK和BNIP3mRNA及其相關(guān)蛋白在52例子宮內(nèi)膜癌組織、14例子宮內(nèi)膜不典型增生組織及36例正常子宮內(nèi)膜組織中的表達情況，，分析子宮內(nèi)膜癌組織中二者的表達水平與臨床病理學(xué)特征的關(guān)系，并對SYK與BNIP3在子宮內(nèi)膜癌中的表達情況進行相關(guān)性檢驗。 結(jié)果：1.在正常子宮內(nèi)膜組、不典型增生組、子宮內(nèi)膜癌組中SYK的表達呈現(xiàn)一種明顯的遞減趨勢，其mRNA的相對表達量分別為1.44±0.36、0.98±0.23、0.63±0.20，SYK蛋白在正常子宮內(nèi)膜組的陽性表達率（91.7%）明顯高于子宮內(nèi)膜癌組（32.7%）、不典型增生組（71.4%），差異均有統(tǒng)計學(xué)意義（P0.01）； 2. BNIP3mRAN在正常子宮內(nèi)膜組、不典型增生組、子宮內(nèi)膜癌組中的相對表達量分別為0.82±0.23、1.99±0.59、1.64±0.46，蛋白水平在子宮內(nèi)膜不典型增生組（92.9%）的表達明顯高于正常子宮內(nèi)膜組（8.3%）與子宮內(nèi)膜癌組(86.5%)，三組差異均有統(tǒng)計學(xué)意義（P0.01）； 3. SYK與子宮內(nèi)膜癌的組織分級、臨床分期、淋巴結(jié)轉(zhuǎn)移、肌層浸潤深度有關(guān)(P0.05)，而BNIP3mRNA及蛋白的表達水平與子宮內(nèi)膜癌病人的臨床病理特征無顯著相關(guān)性（P0.05）； 4.子宮內(nèi)膜癌中SYK與BNIP3兩者的表達呈負(fù)相關(guān)。 結(jié)論：SYK與BNIP3的異常表達與子宮內(nèi)膜正常細(xì)胞的惡變、浸潤、轉(zhuǎn)移等均存在一定的相關(guān)性，聯(lián)合檢測SYK及BNIP3的表達情況可能有助于子宮內(nèi)膜惡性變的早期診斷及預(yù)后的判斷。
[Abstract]:Objective: to investigate the expression and clinical significance of spleen tyrosine kinase (Spleen tyrosine kinase) and apoptotic mitochondrial protein Bcl-2adenocarcinoma E1B19kD interacting protein 3 (BNip3) in endometrial carcinoma, endometrial atypical hyperplasia and normal endometrium.Methods: the expression of SYK, BNIP3mRNA and their related proteins in 52 cases of endometrial carcinoma tissues, 14 cases of endometrial atypical hyperplasia and 36 cases of normal endometrial tissues were detected by real-time quantitative PCR and immunohistochemistry.To analyze the relationship between the expression level of SYK and BNIP3 and the clinicopathological characteristics of endometrial carcinoma, and to test the correlation between the expression of SYK and BNIP3 in endometrial carcinoma.The result is 1: 1.The expression of SYK in normal endometrium, atypical hyperplasia and endometrial carcinoma showed a decreasing trend.The relative expression of mRNA was 1.44 鹵0.36 ~ 0.98 鹵0.23 ~ 0.63 鹵0.20 ~ 0.20 in normal endometrium, respectively. The positive expression rate of mRNA in normal endometrium was significantly higher than that in endometrial carcinoma (32.7%), and in atypical hyperplasia (71.4%), the difference was statistically significant (P0.01).2.BNIP3mRAN in normal endometrium group, atypical hyperplasia group,The relative expression of protein in endometrial carcinoma group was 0.82 鹵0.231.99 鹵0.59 鹵1.64 鹵0.46, respectively. The expression of protein in endometrial atypical hyperplasia group was significantly higher than that in normal endometrial carcinoma group (8.3cm) and endometrial carcinoma group (86.5%). There was significant difference among the three groups (P 0.01).3.SYK was related to histological grade, clinical stage, lymph node metastasis and depth of myometrium invasion of endometrial carcinoma, while the expression of BNIP3mRNA and protein had no significant correlation with the clinicopathological features of endometrial carcinoma.4.The expression of SYK and BNIP3 were negatively correlated in endometrial carcinoma.Conclusion the abnormal expression of SYK and BNIP3 may be related to the malignant change, invasion and metastasis of endometrial normal cells. The combined detection of SYK and BNIP3 may be helpful to the early diagnosis and prognosis of endometrial malignancy.
