miR-145和IL-10在子宮內(nèi)膜樣腺癌的表達(dá)及其相關(guān)性研究

【摘要】:背景子宮內(nèi)膜癌是女性生殖系統(tǒng)常見(jiàn)的惡性腫瘤之一,大約占女性生殖道惡性腫瘤的20%~30%,嚴(yán)重威脅婦女的健康、生命和生活質(zhì)量。該病是發(fā)生于子宮內(nèi)膜的一組上皮性惡性腫瘤,以來(lái)源于子宮內(nèi)膜腺體的腺癌最常見(jiàn)。其發(fā)病率及死亡率近年來(lái)逐漸升高,且更趨向于年輕化。該病具體形成過(guò)程不清,可能與細(xì)胞因子、癌基因、抑癌基因、內(nèi)外環(huán)境等多種因素相互作用有關(guān)。微小RNA-145(mi R-145)是一類(lèi)內(nèi)源性非編碼的小分子單鏈RNA,可在轉(zhuǎn)錄水平調(diào)控基因的表達(dá),參與多種生物學(xué)行為,影響腫瘤的發(fā)生、發(fā)展,發(fā)揮著抑癌基因的作用。白細(xì)胞介素-10(IL-10)是具有多種生物學(xué)活性較強(qiáng)的免疫抑制因子,促進(jìn)腫瘤的發(fā)生。目的研究微小RNA-145(mi R-145)和白細(xì)胞介素-10(IL-10)在不同分化程度的子宮內(nèi)膜樣腺癌患者病灶組織中的表達(dá)情況及其差異性,以及mi R-145與IL-10兩者的表達(dá)是否具有相關(guān)性。了解mi R-145與IL-10兩者在子宮內(nèi)膜樣腺癌中的變化及其意義。方法收集2011年6月至2015年8月于青島大學(xué)附屬醫(yī)院因子宮內(nèi)膜樣腺癌手術(shù)的患者65例,所取子宮內(nèi)膜樣腺癌患者病灶的組織標(biāo)本作為實(shí)驗(yàn)組,其中病理結(jié)果為高分化者標(biāo)本21例,中分化者標(biāo)本27例,低分化者標(biāo)本17例。取同期在門(mén)診上刮宮經(jīng)病理證實(shí)為子宮內(nèi)膜單純性增生的患者組織標(biāo)本23例作為對(duì)照組。采用實(shí)時(shí)熒光定量PCR法技術(shù),檢測(cè)mi R-145和IL-10在實(shí)驗(yàn)組及對(duì)照組病理組織標(biāo)本中的表達(dá)量,分析mi R-145與IL-10在子宮內(nèi)膜樣腺癌患者中的表達(dá)差異性及是否具有相關(guān)性。結(jié)果(1)與單純性增生的子宮內(nèi)膜病灶相比,mi R-145在子宮內(nèi)膜樣腺癌病灶中的表達(dá)水平較低,即對(duì)照組高分化者組中分化者組低分化者組,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);(2)在子宮內(nèi)膜樣腺癌病灶組織中,mi R-145的表達(dá)水平隨著病灶分化程度的降低而降低,即高分化者組中分化者組低分化者組,兩者呈正相關(guān)(P0.05);(3)與單純性增生的子宮內(nèi)膜病灶相比,IL-10在子宮內(nèi)膜樣腺癌患者中的表達(dá)水平明顯升高,即對(duì)照組高分化者組中分化者組低分化者組,差異具有顯著統(tǒng)計(jì)學(xué)意義(P0.01);(4)在子宮內(nèi)膜樣腺癌病灶組織中,IL-10的表達(dá)水平隨著病灶分化程度的降低而升高,即高分化者組中分化者組低分化者組,兩者呈負(fù)相關(guān)(P0.01);(5)在子宮內(nèi)膜樣腺癌病灶組織中,mi R-145與IL-10之間呈負(fù)相關(guān)(P0.05)。結(jié)論在子宮內(nèi)膜樣腺癌患者病灶組織中,mi R-145的表達(dá)下降,IL-10的表達(dá)升高,mi R-145與IL-10兩者的表達(dá)呈負(fù)相關(guān)。mi R-145的表達(dá)下降及IL-10的表達(dá)升高,可能與子宮內(nèi)膜樣腺癌的發(fā)生、發(fā)展有一定關(guān)系。機(jī)體可能通過(guò)某種機(jī)制上調(diào)mi R-145表達(dá),下調(diào)IL-10表達(dá)或mi R-145通過(guò)對(duì)IL-10的上調(diào)作用,來(lái)抑制子宮內(nèi)膜細(xì)胞的病變。由此推測(cè),mi R-145及IL-10的異常表達(dá)或許對(duì)子宮內(nèi)膜樣腺癌的發(fā)生、發(fā)展起到一定作用。
[Abstract]:Background endometrial carcinoma is one of the most common malignant tumors in the female reproductive system, accounting for about 20% 30% of the female genital tract malignant tumors, which is a serious threat to women's health, life and quality of life.The disease is a group of epithelial malignant neoplasms occurring in the endometrium. Adenocarcinoma originating from endometrial glands is the most common.Its morbidity and mortality have increased in recent years and tend to be younger.The formation process of the disease is unclear, which may be related to the interaction of cytokines, oncogenes, tumor suppressor genes, internal and external environment and so on.Small RNA-145(mi R-145) is a class of endogenous non-coding single-stranded RNAs that regulate gene expression at the transcriptional level, participate in many biological behaviors, affect the occurrence and development of tumors, and play a role as tumor suppressor genes.Interleukin-10 (IL-10) is an immunosuppressive factor with high biological activity, which promotes tumor development.Objective to study the expression and difference of minimal RNA-145(mi R-145) and interleukin-10mil (IL-10) in endometrioid adenocarcinoma with different differentiation, and whether there is a correlation between the expression of miR-145 and IL-10.To investigate the changes and significance of mi R 145 and IL-10 in endometrial adenocarcinoma.Methods from June 2011 to August 2015, 65 patients with endometrial adenocarcinoma underwent surgery in Qingdao University affiliated Hospital. The tissue specimens of endometrioid adenocarcinoma were collected as experimental group.The pathological results included 21 cases of highly differentiated specimens, 27 cases of moderately differentiated specimens and 17 cases of poorly differentiated specimens.Tissue specimens of 23 patients with endometrial simple hyperplasia confirmed by pathology in outpatient clinic were taken as control group.The expression of miR-145 and IL-10 in pathological tissues of experimental group and control group was detected by real-time fluorescence quantitative PCR technique. The difference and correlation between miR-145 and IL-10 in endometrioid adenocarcinoma were analyzed.Results (1) compared with the endometrial lesions with simple hyperplasia, the expression of omi R-145 was lower in endometrial adenocarcinoma, that is, the control group, the well-differentiated group, the low-differentiated group, the control group, the well-differentiated group, the low-differentiated group, the control group, the middle differentiation group and the low differentiation group.The difference was statistically significant (P 0.05). The expression level of mil R-145 in endometrial adenocarcinoma decreased with the decrease of the degree of differentiation, that is, the middle differentiated group, the low differentiated group, the middle differentiated group, the low differentiated group.The expression of IL-10 in endometrioid adenocarcinoma patients was significantly higher than that in simple proliferative endometrial lesions, that is, the expression of IL-10 in the control group was significantly higher than that in the low-differentiated group of the well-differentiated group, while the expression level of IL-10 in the endometrial carcinoma was significantly higher than that in the simple proliferative endometrial lesion.The expression of IL-10 in endometrial adenocarcinoma tissues increased with the decrease of the degree of differentiation, that is, the expression of IL-10 in the moderately differentiated group was lower than that in the poorly differentiated group.There was a negative correlation between MMI R-145 and IL-10 in endometrial adenocarcinoma.Conclusion the expression of mil R-145 and IL-10 in endometrioid adenocarcinoma is decreased and the expression of IL-10 is negatively correlated with the expression of IL-10. The decrease of expression of miR-145 and the increase of IL-10 may be associated with the occurrence of endometrial adenocarcinoma.Development has something to do with it.The expression of miR-145 may be upregulated, the expression of IL-10 may be down-regulated, or the up-regulation of IL-10 may be induced to inhibit the pathological changes of endometrial cells.It is suggested that the abnormal expression of miR-145 and IL-10 may play a role in the occurrence and development of endometrial adenocarcinoma.
