【摘要】：目的：探討骨橋蛋白(osteopontin,OPN)在宮腔粘連患者子宮內(nèi)膜中的表達(dá)情況及其在宮腔粘連發(fā)病機(jī)制中的作用。 方法：采用免疫組織化學(xué)法(immunohistochemistry,IHC)及逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)(reverse transcription-PCR,RT-PCR)法,分別檢測70例宮腔粘連(intrauterine adhension, IUA)患者子宮內(nèi)膜及30例子宮縱隔患者正常子宮內(nèi)膜組織中OPN的蛋白及mRNA的表達(dá)情況,比較宮腔粘連組與正常組子宮內(nèi)膜OPN的表達(dá)量的差異,以及不同程度宮腔粘連組子宮內(nèi)膜OPN的表達(dá)量的差異,并對結(jié)果進(jìn)行分析。 結(jié)果：1.免疫組織化學(xué)檢測結(jié)果：OPN蛋白主要在子宮內(nèi)膜腺體表達(dá),間質(zhì)有少量表達(dá)。IUA組的表達(dá)高于非IUA組,差異具有統(tǒng)計學(xué)意義(P0.05)；OPN蛋白在重度IUA組的表達(dá)高于中度IUA組,中度IUA組的表達(dá)高于輕度IUA組,差異具有統(tǒng)計學(xué)意義(P0.05)。2.逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)法檢測結(jié)果：OPNmRNA在IUA組的表達(dá)高于非IUA組,差異具有統(tǒng)計學(xué)意義(P0.05)；重度IUA組的表達(dá)高于中度IUA組,中度IUA組的表達(dá)高于輕度IUA組,差異具有統(tǒng)計學(xué)意義(P0.05),與免疫組織化學(xué)檢測結(jié)果相符。 結(jié)論：1.OPN在宮腔粘連患者子宮內(nèi)膜中表達(dá)明顯增強(qiáng),提示OPN可能與宮腔粘連的發(fā)生有關(guān)；2.隨著宮腔粘連程度的加重,OPN的表達(dá)逐漸增強(qiáng),提示OPN可能與宮腔粘連的嚴(yán)重程度相關(guān)。
[Abstract]:Objective: to investigate the expression of osteopontinin (OPN) in endometrium of patients with intrauterine adhesion and its role in pathogenesis of intrauterine adhesion.Methods: the expression of OPN protein and mRNA in endometrium of 70 cases of intrauterine adhesion (IUAA) and 30 cases of normal endometrium of mediastinum were detected by immunohistochemistry and reverse transcription-PCR reverse polymerase chain reaction (RT-PCR).To compare the difference of OPN expression between uterine cavity adhesion group and normal group, and the difference of endometrial OPN expression in different degree of uterine cavity adhesion group, and to analyze the results.The result is 1: 1.The results of immunohistochemistry showed that the expression of OPN protein was mainly in endometrial glands. The expression of P0.05 IUA protein in interstitial tissue was higher than that in non-IUA group. The difference was statistically significant in severe IUA group than in moderate IUA group, and the expression of P0.05% OPN protein in severe IUA group was significantly higher than that in moderate IUA group.The expression of IUA in moderate IUA group was higher than that in mild IUA group, and the difference was statistically significant (P 0.05). 2.The results of reverse transcription-polymerase chain reaction showed that the expression of IUA was higher in IUA group than in non-#en1# group, the difference was statistically significant (P 0.05), the expression of IUA in severe IUA group was higher than that in moderate IUA group, and the expression in moderate IUA group was higher than that in mild IUA group.The difference was statistically significant (P 0.05), which was consistent with the results of immunohistochemistry.Conclusion: 1. The expression of OPN in endometrium of patients with intrauterine adhesion is significantly increased, suggesting that OPN may be related to the occurrence of intrauterine adhesions.With the severity of intrauterine adhesion, the expression of OPN was gradually increased, suggesting that OPN might be related to the severity of intrauterine adhesion.
