環(huán)氧化酶-2基因啟動(dòng)子區(qū)甲基化水平在子宮內(nèi)膜異位癥中的作用

【摘要】:目的檢測(cè)環(huán)氧化酶-2(COX-2)基因啟動(dòng)子區(qū)CRE位點(diǎn)甲基化狀況,探討其在子宮內(nèi)膜異位癥(EMs)發(fā)生中的作用。方法收集南昌大學(xué)第一附屬醫(yī)院40例子宮內(nèi)膜異位癥患者的子宮內(nèi)膜作為病例組,同期40例正常子宮內(nèi)膜作為對(duì)照組,采用焦磷酸鹽測(cè)序技術(shù)檢測(cè)病例組和對(duì)照組子宮內(nèi)膜組織中COX-2基因啟動(dòng)子區(qū)CRE位點(diǎn)甲基化頻率;采用實(shí)時(shí)定量PCR及免疫組化技術(shù)分別檢測(cè)病例組和對(duì)照組子宮內(nèi)膜組織中COX-2 mRNA及蛋白的表達(dá);分析COX-2基因啟動(dòng)子區(qū)CRE位點(diǎn)甲基化頻率與COX-2表達(dá)水平的相關(guān)性。結(jié)果對(duì)照組內(nèi)膜組織COX-2基因啟動(dòng)子區(qū)CRE位點(diǎn)的甲基化頻率(40.81%±6.10%)顯著高于病例組(14.74%±2.84%),P0.05;而對(duì)照組COX-2 mRNA表達(dá)水平(0.98±0.37)及蛋白表達(dá)水平(1.00±0.73)顯著低于病例組的mRNA表達(dá)水平(2.63±0.44)及蛋白表達(dá)水平(2.74±1.18),P均0.05。隨著COX-2基因啟動(dòng)子區(qū)CRE位點(diǎn)甲基化頻率升高,COX-2基因表達(dá)水平下降,二者呈負(fù)相關(guān)(P0.05)。結(jié)論 COX-2基因啟動(dòng)子區(qū)CRE位點(diǎn)去甲基化與EM s患者在位內(nèi)膜COX-2高表達(dá)相關(guān),在EM s的發(fā)生、發(fā)展中具有重要作用,提示子宮內(nèi)膜異位癥是一種表觀遺傳性疾病。
[Abstract]:Objective to detect the methylation of CRE site in the promoter of cyclooxygenase-2 moxigenin (COX-2) gene and to explore its role in the pathogenesis of endometriosis.Methods the endometrium of 40 patients with endometriosis and 40 normal endometrium as control group were collected from the first affiliated Hospital of Nanchang University.The methylation frequency of CRE site in the promoter region of COX-2 gene was detected by pyrophosphate sequencing in the endometrium of the case group and the control group.The expression of COX-2 mRNA and protein in endometrial tissues of case group and control group were detected by real-time quantitative PCR and immunohistochemistry, and the correlation between CRE methylation frequency of COX-2 gene promoter and COX-2 expression was analyzed.緇撴灉瀵圭収緇勫唴鑶滅粍緇嘋OX-2鍩哄洜鍚姩瀛愬尯CRE浣嶇偣鐨勭敳鍩哄寲棰戠巼(40.81%鹵6.10%)鏄捐憲楂樹簬鐥呬緥緇,
