環(huán)氧化酶-2基因啟動子區(qū)甲基化水平在子宮內膜異位癥中的作用

【摘要】:目的檢測環(huán)氧化酶-2(COX-2)基因啟動子區(qū)CRE位點甲基化狀況,探討其在子宮內膜異位癥(EMs)發(fā)生中的作用。方法收集南昌大學第一附屬醫(yī)院40例子宮內膜異位癥患者的子宮內膜作為病例組,同期40例正常子宮內膜作為對照組,采用焦磷酸鹽測序技術檢測病例組和對照組子宮內膜組織中COX-2基因啟動子區(qū)CRE位點甲基化頻率;采用實時定量PCR及免疫組化技術分別檢測病例組和對照組子宮內膜組織中COX-2 mRNA及蛋白的表達;分析COX-2基因啟動子區(qū)CRE位點甲基化頻率與COX-2表達水平的相關性。結果對照組內膜組織COX-2基因啟動子區(qū)CRE位點的甲基化頻率(40.81%±6.10%)顯著高于病例組(14.74%±2.84%),P0.05;而對照組COX-2 mRNA表達水平(0.98±0.37)及蛋白表達水平(1.00±0.73)顯著低于病例組的mRNA表達水平(2.63±0.44)及蛋白表達水平(2.74±1.18),P均0.05。隨著COX-2基因啟動子區(qū)CRE位點甲基化頻率升高,COX-2基因表達水平下降,二者呈負相關(P0.05)。結論 COX-2基因啟動子區(qū)CRE位點去甲基化與EM s患者在位內膜COX-2高表達相關,在EM s的發(fā)生、發(fā)展中具有重要作用,提示子宮內膜異位癥是一種表觀遺傳性疾病。
[Abstract]:Objective to detect the methylation of CRE site in the promoter of cyclooxygenase-2 moxigenin (COX-2) gene and to explore its role in the pathogenesis of endometriosis.Methods the endometrium of 40 patients with endometriosis and 40 normal endometrium as control group were collected from the first affiliated Hospital of Nanchang University.The methylation frequency of CRE site in the promoter region of COX-2 gene was detected by pyrophosphate sequencing in the endometrium of the case group and the control group.The expression of COX-2 mRNA and protein in endometrial tissues of case group and control group were detected by real-time quantitative PCR and immunohistochemistry, and the correlation between CRE methylation frequency of COX-2 gene promoter and COX-2 expression was analyzed.緇撴灉瀵圭収緇勫唴鑶滅粍緇嘋OX-2鍩哄洜鍚姩瀛愬尯CRE浣嶇偣鐨勭敳鍩哄寲棰戠巼(40.81%鹵6.10%)鏄捐憲楂樹簬鐥呬緥緇,
