Snail在卵巢漿液性腺癌中表達及與上皮間質(zhì)轉(zhuǎn)化的關(guān)系

【摘要】:目的: 檢測Snail、E-cadherin及Vimentin在三種不同卵巢組織中的表達,分析三種蛋白在卵巢漿液性腺癌中表達與臨床病理特征的關(guān)系及其三者之間表達強度的相關(guān)性,探討Snail介導的上皮間質(zhì)轉(zhuǎn)化(EMT)在卵巢癌發(fā)生、發(fā)展及轉(zhuǎn)移中的作用。 方法: 本文采用免疫組化法檢測Snail、E-cadherin及Vimentin分別在48例卵巢漿液性腺癌、48例卵巢交界性漿液性腫瘤及48例卵巢漿液性腺瘤中表達情況,探討三種蛋白的表達與卵巢漿液性腺癌FIGO分期、分化級別、淋巴結(jié)轉(zhuǎn)移及腹膜種植之間的關(guān)系,并分析Snail、E-cadherin及Vimentin在卵巢卵巢漿液性腺癌中表達的相關(guān)性。 結(jié)果: 1.Snail在卵巢漿液性腺癌中的陽性表達率為68.75%,高于卵巢交界性漿液性腫瘤組的41.47%和卵巢漿液性腺瘤組的25%,結(jié)果具有統(tǒng)計學意義(P0.05);Vimentin在卵巢漿液性腺癌中的陽性表達率為66.67%,高于卵巢交界性漿液性腫瘤組的37.5%和卵巢漿液性腺瘤組的18.75%,結(jié)果具有顯著地統(tǒng)計學意義(P0.01);E-cadherin在卵巢漿液性腺癌中的陽性表達率為27.08%,與卵巢交界性漿液性腫瘤組的54.17%和卵巢漿液性腺瘤組的75%相比陽性表達率明顯降低,其結(jié)果具有統(tǒng)計學意義(P0.05)。 2.在卵巢漿液性腺癌組織中,Snail、Vimentin在FIGO分期晚(Ⅲ-Ⅳ期)、分化級別高(中-低分化)、發(fā)生淋巴結(jié)及腹膜轉(zhuǎn)移的腫瘤組織中高表達,而在FIGO分期早(Ⅰ-Ⅱ期)、分化級別低(高分化)、無淋巴結(jié)轉(zhuǎn)移及腹膜轉(zhuǎn)移的腫瘤組織中低表達。E-cadherin在FIGO分期晚(Ⅲ-Ⅳ期)、分化級別高(中-低分化)、有淋巴結(jié)轉(zhuǎn)移及腹膜轉(zhuǎn)移的組織中的表達明顯低于FIGO分期早(Ⅰ-Ⅱ期)、分化級別低(高分化)、無淋巴結(jié)及腹膜轉(zhuǎn)移的卵巢癌組織。 3. Spearman相關(guān)分析顯示:Snail與E-cadherin在卵巢漿液性腺癌中的表達呈負相關(guān)(r=-0.601,P0.01);Snail與Vimentin在卵巢漿液性腺癌中的表達呈正相關(guān)(r=0.477,P0.01); E-cadherin與Vimentin在卵巢漿液性腺癌中的表達相關(guān)性不明顯(r=-0.206,P0.05)。 結(jié)論: 在上皮性卵巢癌組織中,轉(zhuǎn)錄因子Snail及間質(zhì)細胞標志物Vimentin表達上調(diào),而上皮細胞標志物E-cadherin表達下調(diào),提示Snail介導的EMT可能與卵巢癌的發(fā)生、發(fā)展及浸潤轉(zhuǎn)移有關(guān)。
[Abstract]:Objective:The expression of Snail-E-cadherin and Vimentin in three kinds of ovarian tissues was detected, and the relationship between the expression of three proteins in ovarian serous adenocarcinoma and the clinicopathological features, and the correlation between the expression intensity of these proteins and the clinicopathological characteristics were analyzed.To investigate the role of Snail-mediated epithelial mesenchymal transformation in the genesis, development and metastasis of ovarian cancer.Methods:The expression of Snail-E-cadherin and Vimentin in 48 cases of ovarian serous carcinoma and 48 cases of ovarian borderline serous tumor and 48 cases of ovarian serous adenoma were detected by immunohistochemical method. The expression of three proteins and the FIGO stage of ovarian serous adenocarcinoma were studied.The relationship among differentiation grade, lymph node metastasis and peritoneal implantation was analyzed. The relationship between Snail-E-cadherin and Vimentin expression in ovarian serous adenocarcinoma was analyzed.Results:The positive expression rate of 1.Snail in ovarian serous adenocarcinoma was 68.75, which was higher than that in borderline ovarian serous tumor (41.47%) and ovarian serous adenoma (25%).66.67% was higher than that in borderline ovarian serous tumor group (37.5%) and ovarian serous adenoma group (18.75%). The positive expression rate of E-cadherin in ovarian serous adenocarcinoma was 27.08%.The positive expression rate of 54.17% was significantly lower than that of ovarian serous adenoma group (75%).The results were statistically significant (P 0.05).2.In ovarian serous adenocarcinoma tissues, Vimentin was highly expressed in tumor tissues with late FIGO stage (stage 鈪,
