【摘要】：目的 檢測(cè)宮頸正常組織、宮頸上皮內(nèi)瘤變3級(jí)(cervical intraepithelial neoplasias,CIN3)及宮頸癌組織中人類黑色素瘤相關(guān)抗原A3(MAGE-A3)的表達(dá),分析該基因在各組織中的變化與臨床意義,并探討該基因在宮頸癌發(fā)生發(fā)展及轉(zhuǎn)移過程中的作用及分子機(jī)制。方法 取2015年1月至2016年4月河北大學(xué)附屬醫(yī)院采集的72例宮頸組織,病理切片診斷患者宮頸組織類型,判斷分化程度,應(yīng)用實(shí)時(shí)熒光定量PCR(q-PCR)和Western印跡法檢測(cè)不同宮頸組織中MAGE-A3基因及其蛋白的表達(dá)水平。結(jié)果 q-PCR結(jié)果提示,宮頸癌組MAGE-A3基因表達(dá)量明顯高于正常宮頸組及CIN3組,三組間差異均有統(tǒng)計(jì)學(xué)意義(P0.05);Western印跡檢測(cè)MAGE-A3蛋白在正常宮頸組織中的陽性表達(dá)率為20%,在CIN3中為36.7%,在宮頸癌(Ⅰ期、ⅡA期)組織中為61.1%,三組間差異有統(tǒng)計(jì)學(xué)意義(P=0.002);其中ⅡA期MAGE-A3蛋白表達(dá)率明顯高于Ⅰ期的宮頸癌組織(P=0.038)。MAGE-A3蛋白在宮頸癌組織中的表達(dá)與病理類型、腫瘤大小、絕經(jīng)狀態(tài)均無明顯相關(guān)性(P0.05)。結(jié)論 MAGE-A3的表達(dá)與宮頸癌的分化級(jí)別、臨床分期及有無淋巴結(jié)轉(zhuǎn)移情況呈正相關(guān)趨勢(shì),該基因可能在宮頸癌的發(fā)生發(fā)展及轉(zhuǎn)移過程中起著重要的作用。
[Abstract]:Objective to detect the expression of human melanoma associated antigen A3 (MAGE-A3) in normal cervical tissues, cervical intraepithelial neoplasia grade 3 (grade 3) and cervical carcinoma tissues, and to analyze the changes and clinical significance of this gene in all tissues. To explore the role and molecular mechanism of the gene in the development and metastasis of cervical cancer, 72 cases of cervical tissues collected from the affiliated Hospital of Hebei University from January 2015 to April 2016 were collected, and the pathological sections were used to diagnose the cervical tissue types of the patients. The expression of MAGE-A3 gene and its protein in different cervical tissues were detected by real-time fluorescence quantitative PCR q-PCR and Western blotting. Results the results of q-PCR showed that the expression of MAGE-A3 gene in cervical cancer group was significantly higher than that in normal cervix group and CIN3 group.
