TSLP和NF-κB在子宮腺肌病子宮內(nèi)膜細(xì)胞中的表達(dá)及意義

【摘要】:目的研究TSLP、TSLPR和NF-κB三種蛋白在子宮腺肌病患者子宮內(nèi)膜腺上皮細(xì)胞和間質(zhì)細(xì)胞中的表達(dá)及血清TSLP水平,探討TSLP、TSLPR和NF-κB在子宮腺肌病發(fā)病機(jī)制中的作用。 方法采用免疫組織化學(xué)SABC法檢測(cè)35例子宮腺肌病患者異位、在位內(nèi)膜及20例子宮肌瘤患者子宮內(nèi)膜腺上皮細(xì)胞和間質(zhì)細(xì)胞中TSLP、TSLPR和NF-κBp65的表達(dá),并分別對(duì)TSLP和NF-κBp65,TSLP和TSLPR兩組蛋白表達(dá)量進(jìn)行相關(guān)性分析;采用ELISA雙抗體夾心法檢測(cè)患者血清TSLP的水平,子宮肌瘤患者和健康體檢者各20例作為對(duì)照。采用Image-proPlus5.1圖像分析系統(tǒng)對(duì)圖像進(jìn)行分析,用SPSS17.0統(tǒng)計(jì)軟件進(jìn)行數(shù)據(jù)處理。 結(jié)果子宮腺肌病患者異位、在位子宮內(nèi)膜腺上皮細(xì)胞和間質(zhì)細(xì)胞中TSLP、TSLPR和NF-κBp65三種蛋白均陽(yáng)性表達(dá),且表達(dá)量明顯高于對(duì)照組(P0.01),異位內(nèi)膜腺上皮細(xì)胞和間質(zhì)細(xì)胞中TSLP、TSLPR和NF-κBp65表達(dá)量均高于在位內(nèi)膜相應(yīng)蛋白的表達(dá)量(P0.01),異位內(nèi)膜腺上皮細(xì)胞和間質(zhì)細(xì)胞中TSLP和NF-κBp65的表達(dá)呈正相關(guān),而子宮腺肌病患者異位、在位子宮內(nèi)膜腺上皮細(xì)胞和間質(zhì)細(xì)胞中的TSLP和TSLPR的表達(dá)均無(wú)明顯的相關(guān)性,,子宮腺肌病患者血清中TSLP的水平高于子宮肌瘤患者(P0.05)和健康體檢者(P0.05)。 結(jié)論(1)子宮腺肌病患者,在位內(nèi)膜TSLP、TSLPR和NF-κB的表達(dá)高于對(duì)照組,表明子宮腺肌病患者在位內(nèi)膜里有不同于正常內(nèi)膜的特質(zhì),這也體現(xiàn)和證明了“在位內(nèi)膜決定論”的觀點(diǎn);(2)子宮腺肌病患者異位內(nèi)膜TSLP、TSLPR和NF-κB表達(dá)顯著高于在位內(nèi)膜的表達(dá),這提示子宮腺肌病患者異位灶的形成可能與TSLP、TSLPR和NF-κB過(guò)表達(dá)有關(guān)。TSLP與TSLPR的作用可能通過(guò)延遲子宮內(nèi)膜細(xì)胞的凋亡或通過(guò)自分泌作用促進(jìn)異位子宮內(nèi)膜細(xì)胞的增殖,造成異位灶的增殖和侵襲粘附作用;TSLP與TSLPR的作用通過(guò)作用于靶細(xì)胞產(chǎn)生炎癥細(xì)胞因子,間接地促進(jìn)Th2型細(xì)胞免疫從而導(dǎo)致局部炎癥反應(yīng)的發(fā)生,促進(jìn)異位病灶的發(fā)展;(3)子宮腺肌病患者異位內(nèi)膜腺上皮細(xì)胞和間質(zhì)細(xì)胞中TSLP和NF-κB的表達(dá)呈正相關(guān),異位內(nèi)膜TSLP的高表達(dá)有可能與NF-κB的激活和過(guò)表達(dá)相關(guān)。
[Abstract]:Objective to investigate the expression of TSLP- TSLPR and NF- 魏 B in endometrial glandular epithelial cells and stromal cells and the level of serum TSLP in patients with adenomyosis, and to explore the role of TSLP- TSLPR and NF- 魏 B in the pathogenesis of adenomyosis. Methods the expression of TSLP- TSLPR and NF- 魏 Bp65 in endometrial epithelial cells and stromal cells of 35 patients with adenomyosis, eutopic endometrium and 20 patients with uterine leiomyoma were detected by immunohistochemical SABC method. The expression of TSLP and NF- 魏 Bp65 TSLP and TSLPR were analyzed respectively, and the serum TSLP levels were detected by ELISA double antibody sandwich method. 20 patients with uterine leiomyoma and 20 healthy controls were analyzed by Image-proPlus5.1 image analysis system, and the data were processed by SPSS17.0 software. Results TSLP- TSLPR and NF- 魏 Bp65 were positive in ectopic, eutopic endometrial glandular epithelial cells and stromal cells in patients with adenomyosis. The expression of TSLPnTSLPR and NF- 魏 Bp65 in ectopic endometrial glandular epithelial cells and stromal cells were significantly higher than those in eutopic endometrium. The expression of TSLP and NF- 魏 Bp65 in ectopic endometrial epithelial cells and stromal cells were positively correlated. However, there was no significant correlation between the expression of TSLP and TSLPR in ectopic, eutopic endometrial epithelial cells and stromal cells in patients with adenomyosis. The level of TSLP in serum of patients with adenomyosis was higher than that of patients with uterine leiomyosis (P0.05) and that of healthy controls. Conclusion (1) the expression of TSLP TSLPR and NF- 魏 B in eutopic endometrium in patients with adenomyosis is higher than that in control group, indicating that the eutopic endometrium of adenomyosis is different from normal endometrium. The expression of TSLP TSLPR and NF- 魏 B in ectopic endometrium was significantly higher than that in eutopic endometrium. These results suggest that the formation of ectopic foci in patients with adenomyosis may be related to the overexpression of TSLP TSLPR and NF- 魏 B. TSLP and TSLPR may promote the proliferation of ectopic endometrial cells by delaying the apoptosis of endometrial cells or by autocrine. The effects of TSLP and TSLPR on the proliferation, invasion and adhesion of ectopic foci can indirectly promote the Th2 type cell immunity and induce local inflammatory response by acting on the target cells to produce inflammatory cytokines. The expression of TSLP and NF- 魏 B in ectopic endometrial glandular epithelial cells and stromal cells was positively correlated with the development of ectopic lesions. The overexpression of TSLP in ectopic endometrium may be related to the activation and overexpression of NF- 魏 B.
