母鼠維生素D缺乏對子代鼠肺發(fā)育及CyclinD1表達(dá)影響的研究

【摘要】:目的觀察大鼠在孕期、哺乳期維生素D(Vitamin D)缺乏對子代鼠的肺組織結(jié)構(gòu)發(fā)育及細(xì)胞周期蛋白D1(CyclinD1)表達(dá)的影響。方法成年雌性SD大鼠完全隨機(jī)分配為維生素D缺乏模型組(VDD組)及正常對照組(對照組),每組共6只,缺D組予完全避光、不含維生素D飼料喂養(yǎng),正常組予正常光照、正常飼料喂養(yǎng),2周后,與成年雄性SD大鼠以2:1比例合籠交配,建立Vit D孕鼠缺乏模型。利用競爭性酶聯(lián)免疫吸附試驗(yàn)技術(shù)(ELISA)檢測母鼠產(chǎn)后21天血清25-(OH)-D3的水平。分別取出生后第1、7、14、21天子代鼠肺組織,光鏡下分別觀察肺組織形態(tài)結(jié)構(gòu),免疫組化法分別檢測肺組織中CyclinD1的表達(dá)。結(jié)果 VDD組雌鼠血清25-(OH)-D3水平顯著低于對照組(P0.05);光鏡下同日齡VDD組肺組織結(jié)構(gòu)較對照組肺泡間隔增寬,肺泡組織少,肺泡小而不規(guī)則,終末細(xì)支氣管及小支氣管形態(tài)不規(guī)則;免疫組化法檢測CyclinD1 VDD組及對照組均在支氣管黏膜上皮細(xì)胞表達(dá)明顯,對照組CyclinD1在生后第1天表達(dá)最強(qiáng),第7、14、21天逐漸減弱,而VDD組CyclinD1在生后第7天表達(dá)最強(qiáng),高于對照組(P0.05),第1天表達(dá)低于對照組(P0.05),第14、21天逐漸減弱,均高于對照組,差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論母鼠孕期、哺乳期維生素D缺乏延緩子代鼠的肺發(fā)育過程,CyclinD1參與肺組織細(xì)胞的增殖,CyclinD1表達(dá)延遲可能是母鼠Vit D缺乏抑制子代鼠肺發(fā)育的可能機(jī)制之一。
[Abstract]:Objective to observe rats during pregnancy, During lactation, vitamin D Vitamin D) deficiency affected lung tissue development and cyclin D 1 expression in offspring rats. Methods Adult female SD rats were randomly assigned to vitamin D deficiency model group (VDD group) and normal control group. (control group, 6 rats in each group), Group D was fed without vitamin D, and group D was fed with normal light. After feeding with normal diet for 2 weeks, adult male Sprague-Dawley rats were mated in the cage at 2:1. A model of Vit D pregnant mice was established. The serum 25-(OH)-D3 levels were detected by competitive enzyme-linked immunosorbent assay (Elisa) on the 21st day postpartum. The lung tissue was taken from the first day of birth and the lung tissue was observed under light microscope. Results the level of serum 25-(OH)-D3 in the VDD group was significantly lower than that in the control group (P 0.05), and the lung tissue structure of the VDD group at the same day of age was wider than that of the control group under light microscope, and the alveolar tissue in the same age group was smaller and irregular than that in the control group. The morphology of terminal bronchioles and bronchioles was irregular, the expression of CyclinD1 in CyclinD1 VDD group and control group was obvious in bronchial epithelial cells, the expression of CyclinD1 in control group was the highest on the 1st day after birth, and gradually decreased on day 7 1421. On the 7th day after birth, the expression of CyclinD1 in the VDD group was the strongest, which was higher than that in the control group (P 0.05). The expression of CyclinD1 in the VDD group was lower than that in the control group on the first day, and gradually decreased on the 14th day after birth, which was higher than that in the control group, and the difference was statistically significant (P 0.05). The delayed expression of cyclin D1 in lung tissue cells may be one of the possible mechanisms by which Vit D deficiency in female mice inhibits the lung development of offspring.
