【摘要】：目的:探討內(nèi)脂素(visfatin,VF)在子癇前期(PE)患者胎盤組織中的表達及意義。方法:選擇2011年8月至2013年12月在溫州醫(yī)科大學附屬第一醫(yī)院住院分娩的孕婦共計100例,根據(jù)病情輕重分為重度PE組、輕度PE組和正常妊娠組。采用蘇木素-伊紅染色法觀察3組胎盤組織病理變化;免疫組化法及real-time PCR技術(shù)分別檢測3組胎盤VF蛋白及mRNA的表達并分析其與子癇前期發(fā)病的關(guān)系。結(jié)果:PE的病理改變主要表現(xiàn)為細胞滋養(yǎng)細胞及合體滋養(yǎng)細胞結(jié)構(gòu)紊亂且形態(tài)不完整;細胞滋養(yǎng)細胞增生,合體滋養(yǎng)細胞結(jié)節(jié)增多;絨毛毛細血管減少、淤血。免疫組化及real-time PCR結(jié)果顯示胎盤組織中內(nèi)脂素定位于合體及細胞滋養(yǎng)層細胞的胞漿,隨著病情的加重,VF蛋白及mRNA表達量逐漸升高,重度PE組明顯高于正常妊娠組,差異有統(tǒng)計學意義(P0.05)。結(jié)論:PE患者存在絨毛血管內(nèi)皮細胞損傷和功能紊亂。胎盤VF蛋白及mRNA在PE孕婦中高表達,表明VF與PE的發(fā)生具有一定關(guān)系。
[Abstract]:Objective: to investigate the expression and significance of visfatin VFF in placenta of preeclampsia patients with PE.Methods: from August 2011 to December 2013, 100 pregnant women were hospitalized and delivered in the first affiliated Hospital of Wenzhou Medical University. According to the severity of the disease, they were divided into three groups: severe PE group, mild PE group and normal pregnancy group. The pathological changes of placenta were observed by hematoxylin eosin staining method. Immunohistochemical method and real-time PCR technique were used to detect the expression of VF protein and mRNA in placenta of three groups and to analyze the relationship between VF protein and preeclampsia. Results the pathological changes of 10% PE were mainly cytotrophoblast and syncytiotrophoblastic structure. Disorder and incomplete form; Cytotrophoblast proliferation, syncytiotrophoblastic nodule increase, chorionic capillary decrease, congestion. Immunohistochemical and real-time PCR results showed that endolipidins in placenta were located in cytoplasm of syncytiotrophoblast and cytotrophoblastic cells. With the exacerbation of the disease, the expression of VF protein and mRNA increased gradually, and the expression of VF protein in severe PE group was significantly higher than that in normal pregnancy group. The difference was statistically significant (P 0.05). Conclusion VF and mRNA are highly expressed in the placenta of pregnant women with PE, suggesting that VF has a certain relationship with the occurrence of PE.
