輸卵管:間充質(zhì)干細(xì)胞的新來源

【摘要】:目的:探討臨床上廢棄的輸卵管組織中是否存在間充質(zhì)干細(xì)胞(MSC)。方法:收取2013年6月至9月在北京海軍總醫(yī)院手術(shù)治療的6例因子宮頸癌、子宮肌瘤、子宮內(nèi)膜癌及卵巢癌等疾病行輸卵管切除術(shù)的輸卵管組織,體外細(xì)胞培養(yǎng)、流式細(xì)胞學(xué)及體外誘導(dǎo)分化鑒定此類細(xì)胞為輸卵管MSC。結(jié)果:體外分離、培養(yǎng)及擴(kuò)增的輸卵管細(xì)胞形態(tài)呈長梭型,細(xì)胞排列均一并呈漩渦狀生長。流式細(xì)胞術(shù)檢測結(jié)果顯示,≥95%的MSC表達(dá)MSC相關(guān)表面標(biāo)記物CD105、CD73和CD90,CD13、CD29、CD44、CD166、HLA-ABC均陽性表達(dá),CD14、CD19、CD34、CD45、HLA-DR均陰性表達(dá)。體外誘導(dǎo)分化顯示,輸卵管干細(xì)胞具有成骨、成脂肪及成軟骨分化的能力;RT-PCR結(jié)果顯示,輸卵管干細(xì)胞經(jīng)誘導(dǎo)分化后,成脂相關(guān)基因AP2,成骨相關(guān)基因Osteopontin及成軟骨相關(guān)基因Sox-9和COMP表達(dá)均陽性。結(jié)論:臨床廢棄的輸卵管組織中存在MSC,提示臨床中廢棄的輸卵管組織是MSC的新來源。
[Abstract]:Objective: to investigate the existence of mesenchymal stem cells (MSCs) in the discarded oviduct tissue. Methods: from June 2013 to September, 6 cases of cervical cancer and uterine leiomyoma were treated in Beijing Naval General Hospital. The oviduct tissue of endometrial carcinoma and ovarian cancer underwent salpingotomy in vitro cell culture flow cytology and differentiation in vitro were used to identify such cells as fallopian tube MSC.Results: in vitro isolation. The cultured and expanded tubal cells showed long fusiform, uniform arrangement and whirlpool growth. The results of flow cytometry showed that, The expression of MSC related surface markers CD105, CD73 and CD90, CD13, CD29, CD44, CD46, CD166T, and HLA-DR were negative in MSC 鈮,
