【摘要】：目的探討控制性超促排卵(COH)周期多囊卵巢綜合征(PCOS)患者生長分化因子-9(GDF-9)、骨形成蛋白-15(BMP-15)的表達(dá)對卵丘細(xì)胞凋亡的影響。方法收集本院行卵胞質(zhì)內(nèi)單精子注射(ICSI)治療的PCOS組(n=30)和對照組(n=60)患者的卵丘細(xì)胞。TUNEL法及流式細(xì)胞術(shù)檢測卵丘細(xì)胞凋亡;免疫組織化學(xué)方法檢測卵丘細(xì)胞GDF-9、BMP-15蛋白表達(dá),運用Motic Med 6.0數(shù)碼醫(yī)學(xué)圖像分析系統(tǒng)對染色強度定量分析;Real-time PCR技術(shù)檢測卵丘細(xì)胞GDF-9、BMP-15、B細(xì)胞淋巴瘤/白血病-2(BCL)-2、BCL-2相關(guān)X蛋白(BAX)mRNA表達(dá)。結(jié)果 PCOS組穿刺卵泡數(shù)、獲卵數(shù)高于對照組(P0.01);PCOS組卵丘細(xì)胞凋亡率高于對照組(P0.05);GDF-9、BMP-15蛋白在PCOS組和對照組卵丘細(xì)胞胞質(zhì)中均呈陽性表達(dá);PCOS患者卵丘細(xì)胞GDF-9、BMP-15、BCL-2、BAX mRNA的表達(dá)低于對照組(P0.05),BAX/BCL-2比值高于對照組(P0.05)。Pearson相關(guān)分析結(jié)果顯示PCOS患者GDF-9、BMP-15與BCL-2 mRNA表達(dá)呈正相關(guān),與BAX mRNA的表達(dá)呈負(fù)相關(guān)。結(jié)論 PCOS患者卵丘細(xì)胞GDF-9、BMP-15異常表達(dá)可能是卵丘細(xì)胞凋亡增加的原因之一。
[Abstract]:Objective to investigate the effect of the expression of growth differentiation factor (GDF-9) and bone morphogenetic protein-15 (BMP-15) on the apoptosis of cumulus cells in patients with polycystic ovary syndrome (PCOSs) during the controlled hyperstimulation of ovulation (COH) cycle. The apoptosis of cumulus cells was detected by Tunel method and flow cytometry in PCOS group (n = 30) and control group (n = 60). Immunohistochemical method was used to detect the expression of GDF-9 BMP-15 protein in cumulus cells. Motic Med 6.0 digital medical image analysis system was used to quantitatively analyze the staining intensity and real-time PCR technique was used to detect the expression of BDF-9BMP-15B cell lymphoma / Leukemia BCL-2BCL-2 associated X protein. Results the number of punctured follicles in the PCOS group was determined. The apoptotic rate of cumulus cells in the control group was higher than that in the control group. The expression of GDF-9BMP-15 protein in the cytoplasm of the cumulus cells in PCOS and control group was higher than that in the control group. The expression of GDF-9BMP-15 BCL-2BAX mRNA in the cumulus cells of PCOS patients was lower than that in the control group (P 0.05) and the ratio of Bax / BCL-2 in the control group was higher than that in the control group. The expression of BMP-15 in patients with PCOS was positively correlated with the expression of BCL-2 mRNA. Conclusion the abnormal expression of BMP-15 in cumulus cells of PCOS patients may be one of the reasons for the increase of apoptosis of cumulus cells.
