GPx4基因多態(tài)性與子癇前期遺傳易感性的相關(guān)性研究

【摘要】:目的子癇前期是一種妊娠期特發(fā)的疾病,嚴(yán)重威脅著孕產(chǎn)婦和胎兒的生命安全。研究發(fā)現(xiàn)氧化應(yīng)激在其發(fā)生發(fā)展中發(fā)揮重要的作用。谷胱甘肽過氧化物酶(glutathione peroxidase,GPx)4是一種活性很強(qiáng)的抗氧化酶,其水平的下降可能與子癇前期的發(fā)病有相關(guān)性。本研究旨在探討山東漢族女性GPx4基因rs713041位點(diǎn)和rs4807542位點(diǎn)單核苷酸多態(tài)性與子癇前期遺傳易感性的關(guān)系。方法從山東漢族人群中選取1008例已確診的子癇前期孕婦作為病例組,1386例健康足月分娩的孕婦作為對照組,運(yùn)用Taq Man探針熒光定量PCR技術(shù)對兩組孕婦外周血DNA進(jìn)行擴(kuò)增,然后對GPx4基因rs713041位點(diǎn)和rs4807542位點(diǎn)進(jìn)行基因型分析以及等位基因頻率分析。結(jié)果(1)GPx4基因rs713041位點(diǎn)的基因型和等位基因頻率在病例組和對照組之間有統(tǒng)計(jì)學(xué)差異(基因型:χ2=12.292,p=0.002;等位基因:χ2=11.035,p=0.001,OR=1.216,95%CI 1.084 1.365)。另外,當(dāng)我們把rs713041位點(diǎn)的病例組和對照組分別分成(CC+CT)亞組和TT亞組后,發(fā)現(xiàn)這兩個亞組的基因型頻率在病例組和對照組之間有統(tǒng)計(jì)學(xué)差異(χ2=11.241,p=0.001,OR=1.417,95%CI 1.155 1.738)。并且,該位點(diǎn)多態(tài)性可能與輕度子癇前期、重度子癇前期以及早發(fā)型子癇前期發(fā)病均有關(guān)聯(lián)。(2)然而,GPx4基因rs4807542位點(diǎn)的基因型分布與等位基因頻率在病例組和對照組之間差異無統(tǒng)計(jì)學(xué)意義(基因型:χ2=2.037,p=0.361;等位基因:χ2=0.346,p=0.556,OR=0.947,95%CI 0.791 1.134)。結(jié)論本研究數(shù)據(jù)表明山東漢族女性GPx4基因rs713041位點(diǎn)多態(tài)性可能與子癇前期遺傳易感性有關(guān)聯(lián);而該基因另一位點(diǎn)rs4807542位點(diǎn)多態(tài)性可能與子癇前期遺傳易感性無關(guān)聯(lián)。
[Abstract]:Objective preeclampsia is a special disease during pregnancy. Oxidative stress plays an important role in its development. Glutathione peroxidase (glutathione peroxidase) Glutathione peroxidase (Glutathione peroxidase) is a highly active antioxidant enzyme. The aim of this study was to investigate the relationship between the single nucleotide polymorphisms of GPx4 gene rs713041 locus and rs4807542 locus and preeclampsia genetic susceptibility in Shandong Han women. A total of 1008 confirmed preeclampsia pregnant women were selected as the case group and 1386 healthy pregnant women in full term delivery as the control group. Taq Man probe fluorescence quantitative PCR technique was used to amplify peripheral blood DNA in two groups of pregnant women. Genotypic analysis and allelic frequency analysis of rs713041 and rs4807542 loci of GPx4 gene were carried out. Results the genotype and allele frequencies of rs713041 locus of GPx4 gene were significantly different between the case group and the control group (genotype). : 蠂 2 / 12.292P = 0.002; allele: 蠂 ~ 2 / 11.035 / p ~ 0.001 / r = 1.216 / 95 CI 1.084 / 1.365. in addition, When we divided the case group and control group of rs713041 locus into two subgroups: CC) and TT subgroup, we found that the genotype frequencies of the two subgroups were significantly different between the case group and the control group (蠂 ~ 2 / 11.241 / p 0.001 ~ 1.1795 / CI 1.155 / 1.738). Moreover, there was a significant difference between the two subgroups in the frequency of genotype between the two subgroups (蠂 ~ 2 / 11.241 / p 0.001). This locus polymorphism may be associated with mild preeclampsia, There was no significant difference in genotype distribution and allele frequency of rs4807542 locus of GPx4 gene between case group and control group. : 蠂 ~ 2 / 0.346P ~ (0.556) ORA 0.947 / 95 CI 0.791 1.134Conclusion the data of this study suggest that the polymorphism of GPx4 gene rs713041 loci may be associated with preeclampsia genetic preeclampsia susceptibility in Shandong Han women. The polymorphism of rs4807542 locus at another locus of this gene may not be associated with preeclampsia genetic susceptibility.
