【摘要】：目的:分析神經(jīng)營(yíng)養(yǎng)素(NTs)家族成員及其酪氨酸激酶受體(Trk)在卵巢癌中的表達(dá)情況及其與患者總生存期(OS)的關(guān)系,評(píng)價(jià)其預(yù)后價(jià)值。方法:利用KaplanMeier plotter(KM plotter)在線數(shù)據(jù)庫(kù)收集卵巢癌患者病理信息,分析NTs家族成員(NGF、BDNF、NT-3、NT-5)及其受體Trk(TrkA、TrkB、TrkC)表達(dá)與卵巢癌患者預(yù)后及臨床病理參數(shù)的相關(guān)性。收集新鮮卵巢癌組織和癌旁組織各6例,Western blot法檢測(cè)NTs及Trk蛋白表達(dá)。結(jié)果:BDNF mRNA高表達(dá)與卵巢癌患者較好的總生存期(OS)相關(guān)(危險(xiǎn)比HR=0.78,95%CI為0.68~0.89,P0.001);NGF、NT-3、NT-5及受體TrkA、TrkB、TrkC表達(dá)與卵巢癌患者的總生存期(OS)無(wú)關(guān)(P0.05)。BDNF mRNA表達(dá)與臨床分期Ⅰ、Ⅱ及Ⅲ期、病理分級(jí)Ⅱ級(jí)、組織學(xué)分型漿液性癌及TP53野生型的卵巢癌患者的OS密切相關(guān)(P0.05)。NGF、NT-3及TrkA表達(dá)均與卵巢癌患者的臨床病理參數(shù)不相關(guān)(P0.05)。NT-5表達(dá)與病理分級(jí)Ⅱ級(jí)的卵巢癌患者的OS相關(guān)(P0.05)。TrkB及TrkC表達(dá)與臨床分期Ⅲ期的卵巢癌患者的OS密切相關(guān)(P0.05)。Western blot法檢測(cè)顯示,NTs家族中NGF、BDNF、NT-5蛋白在卵巢癌組織中過(guò)表達(dá),NT-3蛋白則為低表達(dá);Trk家族中TrkA、TrkB及TrkC在卵巢癌組織中高表達(dá)。結(jié)論:BDNF表達(dá)與卵巢癌患者的預(yù)后密切相關(guān),高表達(dá)患者的預(yù)后更好,可作為潛在的預(yù)后指標(biāo)進(jìn)行研究;NTs家族其他成員及受體Trk家族均無(wú)此預(yù)測(cè)作用。
[Abstract]:Objective: to investigate the expression of neurotrophin NTs) family and its tyrosine kinase receptor (Trk) in ovarian cancer and its relationship with total survival time (OS). Methods: KaplanMeier plotter(KM plotter-based online database was used to collect pathological information of ovarian cancer patients. To analyze the correlation between the expression of NTs family member BDNFNT-3 NT-5) and its receptor TrktkAK B (TrkBU TrkC5) and the prognosis and clinicopathological parameters of ovarian cancer. The expression of NTs and Trk protein were detected by Western blot in 6 fresh ovarian cancer tissues and 6 paracancerous tissues respectively. Results the expression of NTs and Trk protein was high in the tissues of fresh ovarian cancer and paracancerous tissues. There was no correlation between the expression of mRNA and clinical stage 鈪，
