乳胞素聯(lián)合卡鉑對卵巢癌SKOV3細胞的增殖抑制作用和促凋亡作用

【摘要】:目的:探討蛋白酶體抑制劑乳胞素(LAC)對體外卵巢癌SKOV3細胞增殖和凋亡的影響,并闡明其作用機制。方法:體外培養(yǎng)卵巢癌SKOV3細胞,采用0、2.5、5.0、10.0和20.0μmol·L~(-1) LAC干預(yù)SKOV3細胞48h,5μmol·L~(-1) LAC干預(yù)SKOV3細胞0、24、48和72h;將細胞分為對照組(細胞不經(jīng)藥物干預(yù))、LAC組(加入5μmol·L~(-1) LAC)、卡鉑組(加入10、20、40及80μmol·L~(-1)卡鉑)和LAC聯(lián)合卡鉑組(加入5μmol·L~(-1) LAC和10、20、40、80μmol·L~(-1)卡鉑)。采用MTT法和流式細胞術(shù)(FCM)檢測各組卵巢癌SKOV3細胞的增殖抑制率和凋亡率。結(jié)果:MTT檢測,隨著LAC作用時間的延長和濃度的升高,SKOV3細胞的增殖受到明顯抑制,48h時LAC的半數(shù)抑制濃度(IC50)為5.36μmol·L~(-1);LAC聯(lián)合卡鉑組細胞的增殖抑制率較相同濃度卡鉑組明顯升高(P0.05),卡鉑的IC50由58.08μmol·L~(-1)下降至18.37μmol·L~(-1)。FCM檢測,隨著LAC作用時間的延長,SKOV3細胞凋亡率呈升高趨勢;與卡鉑組和LAC組比較,LAC聯(lián)合卡鉑組SKOV3細胞凋亡率明顯升高(P0.05)。結(jié)論:LAC可有效抑制卵巢癌SKOV3細胞的體外增殖且能誘導(dǎo)其凋亡,并可以明顯增強卡鉑對卵巢癌SKOV3細胞株的增殖抑制作用。
[Abstract]:Objective: to investigate the effects of lactobacillus inhibitor (Lactadine) on proliferation and apoptosis of ovarian cancer SKOV3 cells in vitro and to elucidate its mechanism. Methods: ovarian cancer SKOV3 cells were cultured in vitro. #number0# 渭 mol 路L ~ (-1) LAC was used to treat SKOV3 cells for 48 h and 5 渭 mol 路L ~ (-1) LAC) LAC was used to treat SKOV3 cells for 48 h and 72 h, respectively, and the cells were divided into two groups: control group (cells without drug intervention) with LACU (5 渭 mol 路L ~ (-1)), carboplatin group (10 渭 mol 路L ~ (-1)) and LAC combined with carboplatin group (5 渭 mol 路L ~ (-1)) LAC and capriplatin group (added 5 渭 mol 路L ~ (-1)) and capplatin group (add 10 渭 mol 路L ~ (-1)) and LAC combined with carboplatin group (5 渭 mol 路L ~ (-1)) and 5 渭 mol 路L ~ (-1) LAC. The proliferation inhibition rate and apoptosis rate of ovarian cancer SKOV3 cells in each group were detected by MTT and flow cytometry. When the proliferation of SKOV3 cells was significantly inhibited with the prolongation of the time and concentration of LAC, the IC50 of LAC was 5.36 渭 mol 路L ~ (-1) and carboplatin was 5.36 渭 mol 路L ~ (-1) + carboplatin. The proliferation inhibition rate of SKOV3 cells was significantly higher than that of the same concentration of carboplatin. IC50 decreased from 58.08 渭 mol 路L ~ (-1) to 18.37 渭 mol 路L ~ (-1) ~ (-1). Compared with carboplatin group and LAC group, the apoptosis rate of SKOV3 cells increased significantly with the prolongation of LAC. Conclusion: LAC can effectively inhibit the proliferation of ovarian cancer SKOV3 cells in vitro and induce apoptosis. And the inhibitory effect of carboplatin on the proliferation of ovarian cancer SKOV3 cell line was significantly enhanced.
