【摘要】：目的:檢測妊娠早期稽留流產(chǎn)患者絨毛組織中HIF-1α、BNIP3、LC3的表達(dá),探討滋養(yǎng)層細(xì)胞自噬異常與稽留流產(chǎn)的關(guān)系。方法:收集正常早孕婦女及稽留流產(chǎn)婦女絨毛組織各30例,采用實(shí)時(shí)熒光定量PCR法檢測HIF-1α、BNIP3 mRNA的表達(dá);采用免疫組化SP法及蛋白印跡法檢測HIF-1α、BNIP3及LC3蛋白的表達(dá)。結(jié)果:兩組絨毛組織中均可見HIF-1α、BNIP3及LC3蛋白的表達(dá),HIF-1α主要定位于滋養(yǎng)細(xì)胞胞核及胞質(zhì),BNIP3及LC3主要定位于滋養(yǎng)層細(xì)胞胞質(zhì)中。與正常早孕組相比,稽留流產(chǎn)組HIF-1α與BNIP3 mRNA和蛋白表達(dá)水平以及LC3Ⅱ/Ⅰ均明顯下降(P0.05)。結(jié)論:絨毛組織滋養(yǎng)層細(xì)胞中HIF-1α/BNIP3信號(hào)通路調(diào)節(jié)的低氧誘導(dǎo)自噬水平下降可能是稽留流產(chǎn)發(fā)生的重要機(jī)制。
[Abstract]:Objective: to detect the expression of HIF-1 偽 BNIP3LC3 in the chorionic villi of patients with missed abortion in early pregnancy, and to explore the relationship between abnormal autophagy of trophoblastic cells and missed abortion. Methods: 30 cases of normal early pregnant women and 30 cases of women with missed abortion were collected. The expression of HIF-1 偽 -BNIP3 mRNA was detected by real-time fluorescence quantitative PCR.
