【摘要】：目的探討子宮內(nèi)膜癌中淀粉樣肽前體樣蛋白2(APLP2)的表達(dá),分析其與各臨床參數(shù)的相關(guān)性。方法通過數(shù)據(jù)挖掘,分析335例腫瘤基因圖譜(TCGA)數(shù)據(jù)庫中的子宮內(nèi)膜癌RNA-SEQ數(shù)據(jù)并結(jié)合患者的完整生存期信息,采用Kaplan-Meier法分析APLP2表達(dá)與子宮內(nèi)膜癌患者生存之間的關(guān)系。采用real-time PCR技術(shù)檢測正常子宮內(nèi)膜和子宮內(nèi)膜癌各30例冰凍組織中APLP2 mRNA的表達(dá)情況。應(yīng)用免疫組織化學(xué)法檢測88例子宮內(nèi)膜癌組織和32例正常子宮內(nèi)膜組織中APLP2蛋白的表達(dá)。結(jié)果 Kaplan-Meier生存曲線分析表明,APLP2低表達(dá)的子宮內(nèi)膜癌患者整體生存率明顯降低(HR=2.87,P=0.002)。實時熒光定量PCR檢測結(jié)果提示,APLP2 mRNA在子宮內(nèi)膜癌組織中表達(dá)明顯低于正常內(nèi)膜組織(P=0.031)。免疫組織化學(xué)結(jié)果顯示:APLP2蛋白在子宮內(nèi)膜癌組織中的陽性表達(dá)率明顯低于正常子宮內(nèi)膜組織(P=0.025),其表達(dá)水平與雌激素受體表達(dá)水平相關(guān)(P=0.010)。結(jié)論 APLP2在子宮內(nèi)膜癌中起到抑癌基因的功能。APLP2基因可能通過雌激素受體途徑起作用。APLP2可能成為子宮內(nèi)膜癌預(yù)后預(yù)測及治療的靶點。
[Abstract]:Objective to investigate the expression of amyloid peptide precursor protein 2 (APLP2) in endometrial carcinoma and analyze its correlation with clinical parameters. To analyze the RNA-SEQ data of endometrial carcinoma in TCGA database of 335 cases of tumor, and combine the complete survival information of patients. The relationship between the expression of APLP2 and the survival of endometrial carcinoma was analyzed by Kaplan-Meier. The expression of APLP2 mRNA was detected by real-time PCR technique in 30 frozen tissues of normal endometrium and 30 cases of endometrial carcinoma. Immunohistochemistry was used to detect the expression of APLP2 mRNA in 30 cases of normal endometrium and 30 cases of endometrial carcinoma. The expression of APLP2 protein was detected in 88 cases of endometrial carcinoma and 32 cases of normal endometrial tissue. Results Kaplan-Meier survival curve analysis showed that the overall survival rate of endometrial carcinoma patients with low expression of APLP2 was significantly lower than that of normal endometrial tissues. Real-time fluorescence quantitative analysis showed that the overall survival rate of patients with endometrial carcinoma with low expression of APLP2 was significantly lower than that of patients with endometrial carcinoma. The expression of APLP2 mRNA in endometrial carcinoma tissues was significantly lower than that in normal endometrial tissues (P < 0.031). Immunohistochemical results showed that the positive expression rate of WAPLP2 protein in endometrial carcinoma was significantly lower than that in normal endometrial tissues. Conclusion APLP2 may play a role as a tumor suppressor gene in endometrial carcinoma. APLP2 gene may act through estrogen receptor pathway. APLP2 may be a prognostic preconditioning of endometrial carcinoma. The target of detection and treatment.
