【摘要】：目的研究維生素E對妊娠期糖尿病小鼠氧化應激反應及炎癥因子的影響。方法將C57BL/6J孕鼠隨機分為3組:空白對照組、妊娠期糖尿病(GDM)模型組、維生素E干預組,模型組和干預組腹腔注射0.25%鏈脲佐菌素(STZ)60 mg/kg造模,隨后干預組灌胃維生素E 5 mg/(kg·d),其余兩組給予等量生理鹽水,連續(xù)7天,定時監(jiān)測體重及血糖,實驗結(jié)束后處死小鼠,檢測血清氧化應激指標谷胱甘肽過氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、丙二醛(MDA)和炎癥因子TNF-α、IL-6、IL-8水平。結(jié)果維生素E可上調(diào)GDM小鼠胰島素水平(P0.05),但未改善血糖水平,提高SOD和GSH-Px活性、降低MDA水平(P0.05),降低TNF-α、IL-6、IL-8水平(P0.05)。結(jié)論維生素E可通過改變GDM小鼠體內(nèi)的氧化應激內(nèi)環(huán)境和降低炎癥水平來改善葡萄糖調(diào)節(jié)能力和胰島素水平。
[Abstract]:Objective to study the effects of vitamin E on oxidative stress and inflammatory factors in gestational diabetic mice. Methods pregnant mice were randomly divided into three groups: blank control group, GDM model group and vitamin E intervention group. The model group and the intervention group were injected intraperitoneally with 0.25% streptozotocin (STZ) for 60 mg/kg, then the intervention group was given intragastric administration of vitamin E 5 mg/(kg 路dl, and the other two groups were given the same amount of normal saline for 7 days. The mice were regularly monitored for body weight and blood glucose, and the mice were killed after the experiment. The levels of GSH-PxD, SOD, MDA, TNF- 偽, IL-6 and IL-8 in serum were measured. Results Vitamin E upregulated the insulin level of GDM mice (P 0.05), but did not improve the blood glucose level and the activities of SOD and GSH-Px. Conclusion Vitamin E can improve glucose regulation ability and insulin level by changing the internal environment of oxidative stress and reducing the level of inflammation in GDM mice.
