維生素B12對(duì)宮內(nèi)發(fā)育遲緩大鼠腦損傷的干預(yù)研究

【摘要】:目的:通過孕鼠孕期低蛋白(6%蛋白)飲食法制造宮內(nèi)發(fā)育遲緩(IUGR)的大鼠腦損傷模型,孕期補(bǔ)充維生素B12,觀察仔鼠體格生長(zhǎng)規(guī)律、腦結(jié)構(gòu)變化及腦組織中髓鞘堿性蛋白(MBP)的動(dòng)態(tài)表達(dá),以探討孕期補(bǔ)充維生素B12對(duì)IUGR仔鼠腦損傷的影響。 方法:選取健康三月齡Sprague-Dawley(SD)大鼠56只,按照雌雄1:1的比例置于同一籠中交配。受孕成功的孕鼠隨機(jī)分為正常對(duì)照組(CC)和IUGR實(shí)驗(yàn)組(S),再按孕期是否補(bǔ)充維生素B12,將實(shí)驗(yàn)組隨機(jī)分為維生素B12干預(yù)組(SV)和非干預(yù)組(SC)。對(duì)照組(CC)母鼠自受孕后第1天開始給予21%正常蛋白飲食,而IUGR組(S)母鼠自受孕后第1天開始給予6%蛋白飲食,建立IUGR模型。同時(shí)維生素B12干預(yù)組(SV)母鼠自受孕后第1天開始給予維生素B12150ug/d灌胃,直至分娩。仔鼠生后由母鼠哺乳喂養(yǎng),均給予母鼠21%正常蛋白飲食喂養(yǎng)、自由飲水。選擇生后1日齡、7日齡、14日齡和21日齡4個(gè)時(shí)間點(diǎn),每個(gè)時(shí)間點(diǎn)每組10只仔鼠,測(cè)量仔鼠體重、頭臀長(zhǎng)及腦濕重,并對(duì)各個(gè)時(shí)間點(diǎn)仔鼠留取腦組織,HE染色觀察海馬結(jié)構(gòu)的變化,免疫組化半定量測(cè)定海馬組織中MBP的表達(dá)水平,取海馬用Westernblot檢測(cè)海馬組織中髓鞘堿性蛋白(MBP)含量的動(dòng)態(tài)表達(dá),同時(shí)檢測(cè)母鼠分娩當(dāng)天及各個(gè)時(shí)間點(diǎn)仔鼠血清中維生素B12(VB12)、同型半胱氨酸(Hcy)的表達(dá)水平。 結(jié)果: 1、三組孕鼠所分娩的仔鼠IUGR發(fā)生率及圍產(chǎn)期死亡率比較:與對(duì)照組(CC)相比,SV、SC組仔鼠IUGR發(fā)生率明顯升高(P0.01),而SC組、SV組仔鼠IUGR發(fā)生率無明顯差異(P0.05)。三組仔鼠圍產(chǎn)期死亡率差異無統(tǒng)計(jì)學(xué)意義(P0.05)。 2、三組仔鼠體重、頭臀長(zhǎng)及腦重變化的動(dòng)態(tài)比較:與對(duì)照組(CC)相比,1日齡IUGR組(SV、SC)仔鼠形體瘦小,,體重、頭臀長(zhǎng)、腦濕重均降低(P0.01),7日齡及14日齡時(shí),SV、SC組體重、頭臀長(zhǎng)、腦濕重仍落后于CC組(P0.05),經(jīng)追趕生長(zhǎng),到21日齡時(shí),SV、SC組體重、頭臀長(zhǎng)與CC組相比無明顯差異(P0.05),但腦濕重仍存在差異(P0.05)。IUGR組中,維生素B12干預(yù)組(SV)仔鼠在1日齡、7日齡、14日齡和21日齡時(shí)體重、頭臀長(zhǎng)、腦濕重均高于非干預(yù)組(SC),但差異并不顯著(P0.05)。 3、三組大鼠海馬組織結(jié)構(gòu)HE染色的變化:1日齡時(shí),與對(duì)照組(CC)相比,IUGR組(S)仔鼠海馬神經(jīng)元細(xì)胞排列疏松、紊亂,部分細(xì)胞水腫明顯,出現(xiàn)空泡,核固縮細(xì)胞比較多,神經(jīng)元細(xì)胞數(shù)目減少。到21日齡時(shí),IUGR組(S)細(xì)胞水腫逐步減輕,但細(xì)胞排列相對(duì)疏松、紊亂,細(xì)胞數(shù)目未見明顯增多。1日齡到21日齡,與SC組比較,SV組仔鼠海馬神經(jīng)元細(xì)胞數(shù)目、出現(xiàn)空泡的細(xì)胞數(shù)目均較少,細(xì)胞排列形態(tài)相對(duì)致密、規(guī)則。 4、三組大鼠海馬組織中髓鞘堿性蛋白(MBP)的表達(dá)變化:三組海馬組織中MBP在1日齡、7日齡時(shí)均無明顯表達(dá),從14日齡時(shí)表達(dá)逐漸增加,21日齡時(shí)最強(qiáng);Western blot檢測(cè)也顯示同樣的表達(dá)趨勢(shì)。同樣,在14日齡和21日齡三組中MBP的表達(dá),SC組表達(dá)最低,其次為SV組,CC組的表達(dá)最高。 5、三組孕鼠分娩后及仔鼠血清中維生素B12(VB12)、同型半胱氨酸(Hcy)水平:三組孕鼠分娩后,SV組VB12水平高于SC、CC組(P<0.01),SC組孕鼠VB12水平低于CC組(P<0.01),而Hcy與VB12的表達(dá)趨勢(shì)相反(P<0.05);隨著日齡漸增加,1日齡到14日齡,各組仔鼠B12水平逐漸降低,Hcy水平逐漸升高,到21日齡時(shí)VB12下降、Hcy上升均不明顯;SV組仔鼠VB12水平高于CC組、SC組(P<0.05),SC組仔鼠VB12水平低于CC組(P<0.05),同時(shí)仔鼠中Hcy與VB12的表達(dá)趨勢(shì)相反(P<0.05)。 結(jié)論: 1、孕期低蛋白(6%)喂養(yǎng)飲食法建立IUGR大鼠腦損傷模型,仔鼠腦重降低,海馬神經(jīng)元細(xì)胞數(shù)目減少、細(xì)胞排列疏松,制模可靠,效果比較理想。 2、IUGR仔鼠生后予正常喂養(yǎng),體格上可實(shí)現(xiàn)追趕生長(zhǎng),但神經(jīng)元數(shù)目、腦重及腦組織MBP的表達(dá)落后于正常仔鼠。 3、孕期補(bǔ)充維生素B12可提高孕鼠及仔鼠體內(nèi)維生素B12水平,降低孕鼠、仔鼠體內(nèi)Hcy水平。 4、孕期補(bǔ)充維生素B12,不能降低宮內(nèi)發(fā)育遲緩發(fā)生率,但有助于宮內(nèi)發(fā)育遲緩仔鼠體重、頭臀長(zhǎng)和腦重的增長(zhǎng),神經(jīng)元細(xì)胞形態(tài)的改善及腦組織MBP的表達(dá),提示維生素B12可能對(duì)宮內(nèi)發(fā)育遲緩腦損傷具有改善作用。
[Abstract]:Objective: the pregnant mice of low protein diet (6% protein) preparation of intrauterine growth retardation (IUGR) in rats with brain injury model, vitamin B12 supplementation during pregnancy, physical observation of rat growth rule, changes in myelin basic protein and brain structure in the brain (MBP) dynamic expression, to explore the effect of vitamin B12 supplementation during pregnancy IUGR damage in neonatal rat brain.
Methods: 3 month old healthy Sprague-Dawley (SD) 56 rats, according to the proportion of male and female 1:1 in the same cage. The pregnant rats were randomly divided into normal control group (CC) and IUGR group (S), according to whether vitamin B12 supplementation during pregnancy, the experimental group were randomly divided into vitamin B12 the intervention group (SV) and non intervention group (SC). The control group (CC) rats from pregnant first days after feeding 21% normal protein diet, and group IUGR (S) rats from first days after conception to give 6% protein diet to establish IUGR model. At the same time vitamin B12 intervention group (SV) from maternal pregnancy first days after giving vitamin B12150ug/d orally, until delivery. The offspring by maternal breastfeeding, mothers were given 21% normal protein diet, drinking water freely. 1 day age after birth, 7 days, 14 days and 21 days 4 time points, each time point of 10 rats in each group rats, rats body measurement Heavy head, rump and brain weight, and each time point for rat brain tissue, to observe the changes of hippocampal structure by HE staining, the expression level of semi quantitative immunohistochemical determination in the hippocampus of MBP and myelin basic protein in hippocampus was detected by Westernblot in hippocampus (MBP) expression of dynamic content, at the same time detection of maternal serum vitamin delivery day and each time point in B12 rats (VB12), homocysteine (Hcy) levels.
