孕期和哺乳期大鼠肝臟膽酸和膽酸調控法尼醇X受體、小異二聚體伴侶受體和膽固醇7α-羥化酶的變化

【摘要】:目的研究健康SD鼠在妊娠和哺乳期肝臟膽酸代謝基因的變化。方法取孕10、14、19d及產(chǎn)后1、7、14、21d的大鼠肝臟,檢測膽酸水平和肝臟法尼醇X受體(FXR)、小異二聚體伴侶受體(SHP)、膽固醇7α-羥化酶(Cyp7α1)的表達。用Western blot法測Cyp7α1的蛋白表達情況。結果孕晚期健康大鼠,肝臟中膽酸及Cyp7α1表達不升高;哺乳期的大鼠,肝臟膽酸增加,FXR表達升高;在孕晚期及哺乳期靶基因SHP表達均高于對照組,且其表達高峰期是第孕19天,其值可達對照組的6倍之多。結論健康大鼠在產(chǎn)后肝臟膽酸合成增加,而孕期減少,可能與Cyp7α1、FXR及SHP表達有關。
[Abstract]:Objective to study the changes of bile acid metabolism genes in the liver of healthy SD rats during pregnancy and lactation. The levels of cholic acid and hepatic FXRX receptor FXRX, small heterodimer chaperone receptor (SHP) were measured. The expression of cholesterol 7 偽 -hydroxylase (Cyp7 偽 1). The expression of Cyp7 偽 1 protein was measured by Western blot method. The expression of cholic acid and Cyp7 偽 1 in liver was not increased. In lactating rats, cholic acid in liver increased the expression of FXR. The expression of target gene SHP in late pregnancy and lactation was higher than that in control group, and the peak expression of target gene was at the 19th day of gestation, which was 6 times higher than that in control group. Conclusion the synthesis of cholic acid in the liver of healthy rats increased after delivery. The decrease in pregnancy may be related to the expression of Cyp7 偽 1 FXR and SHP.
