【摘要】：目的通過檢測接受體外受精-胚胎移植(In vitro fertilization and embryo transfer, IVF-ET)治療的患者外周血白細(xì)胞上卵泡刺激素受體(Follicle-stimulating Hormone Receptor, FSHR)啟子-29位點(diǎn)的基因型,探討該位點(diǎn)基因突變后表達(dá)改變與IVF-ET患者卵巢反應(yīng)性的關(guān)系。 方法本研究根據(jù)在IVF-ET治療周期中,患者在接受控制性超促排卵(Controlled ovarian hyperstimulation, COH)時(shí)卵巢對藥物的反應(yīng)性,將85例患者分為三組,其中卵巢高反應(yīng)組20例,卵巢反應(yīng)正常組46例,卵巢低反應(yīng)組19例。采用聚合酶鏈?zhǔn)椒磻?yīng)-限制性長度多態(tài)性技術(shù)(Polymerase chain reaction-restriction fragment length polymorphism, PCR-RFLP)來測定患者白細(xì)胞上FSHR啟動(dòng)子-29位點(diǎn)的基因型,并分析其與患者卵巢反應(yīng)性的關(guān)系。 結(jié)果①卵巢低反應(yīng)組FSHR啟動(dòng)子29位點(diǎn)G→A突變率為,42.10%,卵巢反應(yīng)正常組的基因突變率為10.87%,卵巢高反應(yīng)組的基因突變率為10%。 ②卵巢低反應(yīng)組的基因突變率均高于卵巢反應(yīng)正常組及高反應(yīng)組(P0.05)；卵巢反應(yīng)正常組和高反應(yīng)組的基因突變率比較無顯著性差異(P0.05)。 ③卵巢低反應(yīng)組HCG日雌二醇(estradiol, E,)值及荻卵數(shù)均低于其他兩組(P0.05)；卵巢反應(yīng)正常組HCG (?) E值及獲卵數(shù)均低于卵巢高反應(yīng)組(P0.05)。 結(jié)論外周血白細(xì)胞FSHR啟動(dòng)子-29位點(diǎn)G→Λ基因突變可能與卵巢反應(yīng)性不良有關(guān),測定該位點(diǎn)的基因型對預(yù)測卵巢的反應(yīng)性有一定的意義。
[Abstract]:Objective to detect in vitro fertilization and embryo transfer. Follicle-stimulating Hormone Receptor was obtained from peripheral white blood cells treated with IVF-ETS. To investigate the relationship between the gene mutation and ovarian reactivity in patients with IVF-ET. Methods according to the treatment cycle of IVF-ET, patients received controlled ovarian hyperstimulation with controlled hyperstimulation of ovulation. 85 patients were divided into three groups: ovarian hyperresponsiveness group (n = 20) and normal ovarian response group (n = 46). Methods Polymerase chain reaction (PCR) and restriction length Polymorphism (RFLP) technique were used in 19 cases of ovarian hyporesponse group (. Polymerase chain reaction-restriction fragment length polymorphism. PCR-RFLP was used to determine the genotype of FSHR promoter-29 on leukocytes and to analyze its relationship with ovarian reactivity. Results 1 FSHR promoter 29 locus G in ovarian hyporesponse group 鈫扵he mutation rate of A was 42.10, the mutation rate of normal ovary was 10.87, and the mutation rate of ovarian high response was 10. 2the gene mutation rate of ovarian low response group was higher than that of normal ovarian response group and high response group (P 0.05). There was no significant difference in gene mutation rate between normal ovarian response group and high response group (P 0.05). 3 the value of estradiol (E) and the number of eggs in the HCG group were lower than those in the other two groups (P 0.05), and the number of estradiol (E) in the normal ovarian response group was lower than that in the other two groups (P 0.05). The value of E and the number of oocytes obtained were lower than that of the ovarian hyperreactive group (P0.05A). Conclusion the FSHR promoter -29 in peripheral blood leukocytes is G 鈫扵he mutation of A gene may be related to ovarian reactivity, and the determination of the genotype of this locus may be useful in predicting ovarian reactivity.
